羧甲基壳聚糖对人肺上皮细胞短路电流影响的研究

摘    要:目的探讨羧甲基壳聚糖对人肺上皮细胞短路电流的影响及其在肺上皮液体转运中的作用。方法应用尤斯灌流室装置测定H441单层细胞的短路电流。由总电流减去阿米洛利可抑制的电流算得阿米洛利敏感性电流,以用药前H441单层细胞的阿米洛利敏感性电流初始值为100%对照。结果羧甲基壳聚糖能够剂量依赖性增加H441单层细胞的短路电流,此电流可被阿米洛利明显抑制;0.1%、0.2%、0.3%羧甲基壳聚糖处理的人H441单层细胞阿米洛利敏感性电流分别增加至(151±16)%、(192±27)%、(222±34)%,与对照值相比P均<0.05。结论羧甲基壳聚糖通过增加人肺上皮细胞阿米洛利敏感性电流而增加肺泡上皮离子转运,可能参与肺水肿液的吸收。

Effects of carboxymethyl chitosan on short-circuit currents in human lung epithelial cells
Abstract:Objective To explore the relationship between carboxymethyl chitosan and short-circuit currents in human lung epithelial cells and to find the role of carboxymethyl chitosan in lung epithelial fluid transport.Methods Short-circuit currents in H441 mono-layers were measured by ussing-chamber technique.The amiloride-sensitive currents were got by the subtraction of amiloride-inhibitable currents from the total currents,and the original value of amiloride-sensitive currents in H441 mono-layers before drug administration was set as 100% control.Results Carboxymethyl chitosan increased the short-circuit currents in H441 monolayers dose-dependently,which could be inhibited obviously by amiloride.0.1% carboxymethyl chitosan could increase the amiloride-sensitive currents to(151±16)%(P<0.05),which showed a significant difference from control.0.2%~0.3% carboxymethyl chitosan could increase the amiloride-sensitive currents to(192±27)% and(222±34)%,respectively(P<0.01),which showed a much significant difference from control.Conclusion Our observations demonstrate that carboxy methyl chitosan can increase the amiloride-sensitive currents in human lung epithelial cells and enhance alveolarion transport,which may be involved in the absorption of lung edema.
