腺苷钴胺凝胶中腺苷钴胺的含量测定

目的 采用高效液相色谱(HPLC)法测定腺苷钴胺凝胶中的腺苷钴胺含量. 方法色谱柱为Uitimate Column XB C₁₈(4.6 mm×250 mm,5 μm),以0.05 mol.L^-1磷酸二氢钾溶液(用磷酸调节pH为3.2)-乙腈(83:17)为流动相;柱温:35 ℃;进样量:10 μL;流速:1.0 mL.min^-1,检测波长:260 nm. 结果 腺苷钴胺在10～50 μg.mL^-1范围内线性关系良好(r＝0.999 9),平均回收率为99.49%,RSD为0.60%(n＝9). 结论 该方法专属性强、准确,可用于测定腺苷钴胺凝胶的含量.

Content Determination of Cobamamide in Cobamamide Gel

Objective To establish a high performance liquid chromatography method for determining cobamamide in cobamamide gel.Methods A XB-C18 Uitimate column(4.6 mm ×250 mm,5 μm)was used;the mobile phase was consisted of 0.05 mol·L-1 monobasic potassium phosphate buffer solution(adjusted to pH 3.2 with phosphoric acid):acetonitrile(83:17);loading volume was 10 μL;the flow rate was 1.0 mL·min-1;and the UV detection wavelength was set at 260 nm.Results The linear range for cobamamide was 10-50 μg·mL-1,r= 0.999 9.The average recovery was 99.49%,with a RSD as 0.60%(n=9).Conclusion The method is specific and accurate,which is suitable for the content determination of cobamamide in the gel.
