小鼠Lewis肺癌移植瘤模型活体分子成像研究

摘    要:目的建立能进行光学活体分子成像的lewis肺癌(LLC)移植瘤模型,探讨其在观察研究肿瘤的生长、转移、肿瘤血管生成及治疗效果等方面的价值。方法利用pLEIN-CMV-GFP逆转录病毒载体将绿色荧光蛋白(green fluorescent protein,GFP)报告基因转入小鼠Lewis肺癌(LLC)肿瘤细胞,体外稳定高效表达后,种植于小鼠双侧腹股沟,在不同时间利用光学活体成像仪成像,观察并用病理学证实。结果所有动物均成功制成荷GFP基因LLC移植瘤模型,GFP基因稳定高效表达,可见随肿瘤生长,绿色荧光团块增大。结论该模型稳定可靠,能够使我们利用光学分子成像的方法,实时、无创、连续地观察肿瘤生长,为研究肿瘤提供了新的思路和方法。

Abstract:Objective To develop an animal model bearing transplanted Lewis lung carcinoma (LLC) that can be examined by molecular imaging,and to study its value in the research of tumor growth,metastasis,and the effect of the treatment.Methods The green fluorescent protein (GFP) gene was transfected into LLC cells by using pLEIN-CMV-GFP vector.After the stable and high expression of GFP,the tumor was subcutaneously transplanted to the groin of the mice and the images were acquired at different periods of time by using optical imaging instrument.At last the result was proved by pathology.Results All the mice in the experiment have been made transplanted tumor models labeled with GFP,and GFP gene was stably and highly expressed.The fluorescence marks grew with the growth of the tumor.Conclusion This animal model with GFP expressed LLC cells for molecular imaging is stable,and relatively uncomplex to make,which provides us a new way with its advantages of real time,noninvasive and in vivo in studying the tumors.
