平帕汤对PD模型小鼠多巴胺能神经元保护作用的研究

摘    要:目的 探讨平帕汤对帕金森病(PD) 模型小鼠多巴胺能神经元的保护作用.方法 在1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP) 诱导的PD 小鼠模型基础上,随机分为正常组,模型组和平帕汤组,采用爬杆测试法观察平帕汤对PD 小鼠行为特征的影响;采用高效液相电化学法(HPLC-ECD) 检测各组小鼠纹状体内多巴胺(DA)、3,4-二羟基苯乙酸(DOPAC) 和高香草酸(HVA) 的含量;采用免疫组化染色法观察各组小鼠黑质酪氨酸羟化酶(TH) 阳性细胞表达的变化.结果 末次注射MPTP 后,与正常组相比,模型组爬杆时间和评分明显增加(P〈0.05),平帕汤组显著增加(P〈0.01) ;与模型组相比,平帕汤组DA、DOPAC 值有上升趋势,DA/HVA 值则明显增加(P〈0.01,P〈0.05) ; 平帕汤组黑质TH 阳性细胞的表达程度明显提高(P〈0.05 或P〈0.01).结论 平帕汤能保护帕金森病模型小鼠脑黑质多巴胺能神经元,挖掘其功能潜力,振奋其分泌能力,从而提高脑内黑质纹状体系统多巴胺的浓度,最终达到防治PD 的作用.

The Research on the Protective Effect of Ping-pa Decoction on Dopaminergic Neurons in PD Mouse Models

Abstract:Objective:To explore the Ping-pa Decoction' s protective effect on dopaminergic neurons in Parkinson' s disease(PD) mouse models.Methods:Based on 1-methyl-4-phenyl-1, 2, 3, 6-tetrahydropyridine (MPTP) induced PD mouse models, they were randomly divided into the normal control, model control and Ping-pa decoction groups.Pole climbing test was used to observe the effect of Ping-pa decoction on characteristics of praxiological change in PD mouse models ; The levels of dopamine (DA), 3, 4-dihydroxyphenylacetic acid (DOPAC) and homovanillic acid(HVA) in striatum were measured by High Performance Liquid Chromatography-Electrochemical Detector(HPLC-ECD).Moreover, immunohistochemical method was used to observe the expression of Tyrosine hydroxylase (TH) in dopaminergic neurons in the substantia nigra.Results:After the last injection of MPTP, compared with the normal control group, the pole climbing time and the score of model cotrol group increased obviously (P〈0.05), Ping-pa decoction group ascended significantly (P〈0.01) ; Levels of DA and the value of DA/DOPAC in Ping-pa decoction group were higher than those in model group, particularly the value of DA/HVA(P〈O.O1,P〈O.05) ; TH expression in the substantia nigra in Ping-pa decoction group was also higher than that of models(P〈0,05 or P〈0.01). Conclusion:Ping-pa decoction can protect dopaminergic neurons in substantia nigra of mice with PD, encourage their functional potential and enhance their DA secretion capacity, thereby increasing the concentration of DA in the nigrostriatal system and ultimately attain the preventive and curative effect of PD.
