黄连解毒汤对糖尿病大鼠脂肪因子visfatin表达的影响

目的 :研究黄连解毒汤对糖尿病大鼠肝脏visfatin表达的影响,探讨黄连解毒汤改善胰岛素抵抗的作用机制。方法 :实验采用高脂高糖饮食联合链脲佐菌素(STZ)复制糖尿病大鼠模型,分为空白组、模型组、对照组、治疗组,连续给药6周后测空腹血糖(FBG)、空腹胰岛素(FINS)、总胆固醇(TC)、三酰甘油(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)含量及肝脏visfatin表达的变化。结果:治疗组血清FBG、FINS、TC、TG、LDL-C水平显著低于模型组(P0.01),HDL-C水平高于模型组(P0.01);治疗组肝脏表达visfatin显著低于模型组(P0.01)。结论:黄连解毒汤能显著降低糖尿病大鼠血脂血糖,改善胰岛素抵抗,其机制可能是调节脂肪因子visfatin的表达,降低炎症信号通路,改善脂肪组织的慢性炎症反应。

Effects of Huanglian Jiedu Decoction on the Expression of Adipocyte Factor Visfatin of Diabetic Rats

Objective：To study effect of Huanglian Jiedu decoction on visfatin expression of diabetes rat liver,in order to discuss the mechanism of improving insulin resistance. Methods：Diabetic rat models were established by high fat and high sugar diet combined with streptozotocin（STZ）. Model rats were divided into blank group,model group,control group,treatment group. After treated with Huanglian Jiedu decoction for 6 weeks, fasting blood glucose （FBG）, fasting insulin （FINS）, total cholesterol （TC）, triglyceride （TG）, high density lipoprotein eholesterol（HDL-C）,low density lipoprotein cholesterol（LDL-C） contents were determined. And the changes of liver visfatin expression were also determined. Results：In Huanglian Jiedu decoction group,the serum levels of FBG,TC,TG,LDL-C,FINS levels were significantly lower than those of the model group（P〈0.01）,the level of HDL-C was higher than that of model group（P〈0.01）,and liver expression of visfatin of the treatment group was significantly lower than that of model group（P〈0.01）. Conelusion：Huanglian Jiedu decoction can significantly reduce the blood sugar of diabetic rats,and improve insulin resistance. The mechanism is probably through regulating the expression of adipocyte factor visfatin,reducing the inflammatory signaling pathway,and reducing the chronic inflammation in adipose tissue.
