夏至草体外抗单纯疱疹病毒作用研究

摘    要:目的 :探索夏至草中不同极性的粗提取物对单纯疱疹病毒I型(HSV-1)的体外抑制作用。并进一步分离纯化,为寻找分离抗HSV-1的活性组分做准备。方法:分别以水、95%乙醇、乙酸乙酯为溶剂提取药物。采用HSV-1-Hep-2细胞体外病毒感染模型,进行体外抗HSV-1作用的评价。将醇提物采用水沉法进一步纯化,分别将上清液和沉淀物进行体外抗病毒实验,通过显微镜下观察细胞病变效应(CPE),以酶标仪测定中性红染色A540值,计算治疗指数(TI)。结果:夏至草的水、乙醇、乙酸乙酯三种总提取物中,乙醇总提取物的抑毒效果最好。夏至草醇提水沉后,上清液的治疗指数TI大于46.5,沉淀物的治疗指数TI大于51.6。结论:夏至草醇总提取物具有抗HSV-1活性,醇提水沉后,其上清液和沉淀物都有显著的抗HSV-1活性。

Abstract:Objective:To explOre crude extracts with different polarity from the Lagopsis supina in vitro inhibiting herpes simplex virus type I (HSV-1),and to do the preparation for isolating the active ingredients for anti-HSV-1 by further purifying. Methods:Pure water,95% ethanol,ethyl acetate were used as the solvent to extract drugs. Hep-2 cells were infected with HSV-1 in vitro to evalute anti-HSV-1 effect. The alcohol extract was further purified by precipitation with water,and sediment and the supernatant were tested for the in vitro antiviral experiments respectively. The cytopathic effect(CPE) was observed under a microscope. Neutral red staining was measured in a microplate reader at 540 nm to calculate the therapeutic iddex(TI). Results:Among the three total extracts from Lagopsis supina,the ethanol total extract is best effect. After alcohol total extract was precipitated with water,the supernatant index T1 was greater than 46.5, and the sediment TI index was greater than 51.6. Conclusion:Total alcohol extract from Lagopsis supina has anti-HSV-1 effects. After processed with ethanol extraction and water precipitation,its supernatant and sediment have obvious anti-HSV-1 effects.
