诱生型一氧化氮合酶在糖尿病早期大鼠肾脏中的表达

摘    要:目的 观察糖尿病早期大鼠肾组织内诱生型一氧化氮合酶(iNOS)表达情况,探讨糖尿病早期大鼠肾组织内iNOS变化的规律与糖尿病肾病的关系.方法应用免疫组织化学链霉菌素亲和生物素--过氧化物酶连接法及图象分析方法(CIAS-1000细胞图象分析系统),测定STZ诱发的糖尿病早期大鼠肾脏内iNOS表达并进行分析.结果(1)1周、2周组iNOS在肾小球部位的表达逐渐上升,且2周为高峰,到4周明显下降.(2)iNOS在近端小管1周、2周、4周均有表达,但无明显峰值变化.结论通过iNOS在糖尿病早期大鼠肾脏(以肾小球中表达的变化较明显)的表达,推测iNOS的表达可能参与糖尿病早期肾损害.

Abstract:Objective To determine whether the variance of inducible nitric oxide(iNOS) is expressed in the kidney of earlier period of diabetic rats as well as the relation of the diabetic kidney disease.Methods The expression of iNOS in the kidneys of STZ induced diabetic rats was examined by immunohistochemitry(Strepavidin Peroxidase conjugated method),using monoclonal goat anti-rat antibody,and analyzed by the portrait analysis method(the cell CIAS-1000 portrait analysis system).Results(1)In 1 week and 2 week groups,iNOS in renal glomerulus was expressed higher and higher,and then in 4 week group showed a significant decrease in contrast to 2 week group.(2)In 1 week,2 week and 4 week groups,iNOS was expressed in proximal convoluted tubule,but the variance of expression was slight.Conclusions Through the variety of expression of iNOS in rats's kidneys of earlier period of diabetes(especially in renal glomerulus),it was maybe iNOS is an important factor in the function injure of diabetes kidney in early days.
