Abstract:Objective:To construct prognostic related competitive endogenous RNA(ceRNA)networks in colon adenocarcinoma(COAD)by using bioinformatics tools and to conduct comprehensive analysis.Methods:RNA sequencing(RNA-seq)data and miRNA sequencing(miRNA-seq)data of COAD were downloaded from The Cancer Genome Atlas(TCGA)database,along with corresponding clinical information of patients,to screen for differentially expressed mRNAs(DEmRNAs),lncRNAs(DElncRNAs)and miRNAs(DEmiRNAs).Function and pathway enrichment analyses,as well as protein-to-protein interaction(PPI)network analyses,were performed for all DEmRNAs.The ceRNA network of COAD was constructed based on the regulatory relationships between these differential RNAs.Kaplan-Meier curves were used to assess the impact of these RNAs on the overall survival(OS)of patients with COAD.The correlation between the expression pattern of differential genes and other clinical features was analyzed in the context of patient-related clinical information.The relationship between DElncRNAs and immune-related molecules andcellswas analyzed using c-bioportal and TIMER database.Results:5 DElncRNAs(C2orf48,HOTAIR,KCNQ1OT1,LINC00355 and MIR31HG),2 DEmiRNAs(miR-145 and miR-152)and 6 DEmRNAs(FAM46C,FJX1,MACC1,SLC16A9,TPM2 and ULBP2)were significantly associated with OS in patients in the ceRNA network constructed by us.In addition,4 of the prognosis-related lncRNAs have been shown to be associated with certain other clinical features.There was a significant correlation between KCNQ1OT1,MIR31HG and PD-L1 expression.Conclusion:This study reveals a novel ceRNA regulatory network for COAD and identified lncRNA C2orf48,HOTAIR,KCNQ1OT1,LINC00355,and MIR31HG as potential biomarkers of COAD for prognosis,targeted therapy,and immunotherapy response.
