内障丸对青春期PCOS大鼠卵巢形态学和血清激素的影响

摘    要:目的:探讨内障丸对青春期PCOS大鼠卵巢形态学和血清激素的影响。方法:以23日龄幼年SD清洁雌性大鼠为研究对象,应用皮下埋植左炔诺孕酮硅胶棒联合皮下注射HCG法建立青春期PCOS动物模型。运用免疫组化及光、电镜技术等方法,检测大鼠造模前后及给药前后卵巢形态学和血清激素的变化。结果:内障丸高、中、低剂量组大鼠用药后体重均明显低于模型组。内障丸治疗后卵巢色泽红润,高剂量组卵巢重量较模型组明显降低。36日模型组卵泡比例较对照组明显增加(P0.01),颗粒细胞层厚度较对照组明显变薄(P0.01),泡膜细胞层厚度较对照组明显增厚(P0.05)。内障丸治疗后高、中、低剂量组囊状卵泡比例均明显降低(P0.01),颗粒细胞层数均较模型组明显增加(P0.05),其中低剂量组黄体组织数量增多明显(P0.05)。造模后模型组E2含量变化不明显,而T含量显著升高(P0.01)。内障丸治疗后高、中剂量组T含量均明显低于模型组(P0.01)。结论:①皮埋左旋18-甲基炔诺酮联合HCC诱导SD幼年雌性大鼠PCOS动物模型卵巢病理改变及血内分泌改变与PCOS病人相似。②内障丸可以改善多囊卵巢大鼠卵巢形态学的异常,其促排卵的机制可能是通过对颗粒细胞和卵泡膜细胞的作用,降低多囊卵巢大鼠雄激素水平,从而促使卵巢排卵。③内障丸明显改善了多囊卵巢大鼠的高雄激素血症,对多囊卵巢大鼠异常的垂体及卵巢局部内分泌环境起到调节作用。
