感觉神经肽在不同程度哮喘时肺组织中含量变化及相关研究

摘    要:目的 研究哮喘豚鼠肺组织中神经激肽A(NKA)、神经激肽B(NKB)及P物质 (SP)在不同严重程度哮喘时含量变化规律及其各神经肽间的相关性 ,探讨其与哮喘发病机制的关系。方法 用卵蛋白超声雾化吸入法制作豚鼠哮喘模型 ,随机分成三组 (每组 15只 )。①哮喘组 (诱喘一次组 ) :卵蛋白致敏后 ,第 2 1天激发诱喘 ;②反复诱喘组 :第 2 1天诱喘后 ,再反复隔日诱喘一次 ,连续一周 ;③正常对照组 :同期雾化吸入生理盐水。研究组豚鼠于诱喘后 (组②于末次诱喘后 ) 2 0~ 2 4h取材 ,肺组织匀浆处理后 ,以酶联免疫方法检测NKA、NKB ;以放射免疫方法检测SP。结果 哮喘组 (诱喘一次 )肺组织中NKA、NKB及SP含量较正常对照组均显著增高 (P均 <0 0 1) ;反复诱喘组肺组织中NKA、NKB及SP含量均明显高于诱喘一次组 ,差异显著 (P <0 0 1或P <0 0 5 ) ;②各组NKA、NKB及SP间均以NKA含量最高 ,显著高于NKB及SP ;诱喘一次及反复诱喘组豚鼠肺组织中NKA与NKB间呈显著正相关 (r =0 74 5 ,P <0 0 5 ) ;NKA与SP间无相关性 (r =0 2 5 3,P >0 0 5 ) ;NKB与SP间亦无相关性 (r=0 15 9,P >0 0 5 )。结论 肺组织中NKA、NKB及SP含量变化 ,与卵蛋白诱发的豚鼠哮喘及病情关系极为密切 ,提示上述感觉神经肽参与了哮喘的发病机制 ,而NKA

Abstract:Objective To study the changes and correlations of neurokinin A(NKA),neurokinin B(NKB) and substance P(SP) in lung tissues of asthmatic guinea with different asthmatic status,and explore the relationship between these neuropeptides and asthma.Methods 45 guinea pigs were randomly divided into normal control group,asthma group and asthma group of repeatedly attacks, 15 guinea pigs for each group. 1.Asthma group:the animals were sensitized by exposure to aerosolized ovalbumin and challenged with the same antigen at the 21d.2. asthma group of repeatedly attacks: the sensitized animals were repeatedly challenged every other day for 1 week after the first challenge with the same antigen at the 21 day.3. Normal control group:the animals were exposure to aerosolized NS in the same time.The animals were anatomized after 24h of the last ovalbumin challenge,the animals of normal contol group were anatomized in the same time.The contents of NKA and NKB in lung tissues were detected by ELISA and the contents of SP in lung tissues were detected by RIA.Results 1.The contents of NKA,NKB and SP in the lung tissues of asthmatic guinea pigs were significantly higher than normal control (P<0.01,respectively),the contents of NKA,NKB and SP of the repeatedly asthmatic guinea pigs were significantly higher than those in asthmatic guinea pigs attacked only once (P<0.01 or P<0.05).2.The contents of NKA was the highest among NKA,NKB and SP in each groups, there was a significantly positive correlation between NKA and NKB in lung tissues of asthmatic guinea pigs (r=0.745,P<0.05) and no significantly correlation between NKA and SP (r=0.253,P>0.05) or NKB and SP(r=0.159,P>0.05).Conclusion The contents of NKA,NKB and SP in the lung tissues have a close relationship with asthma that induced by ovalbumin.NKA,NKB and SP may play an important role in the pathophysiologic process of asthma. NKA is the predominant tachykinin in asthmatic lung tissues.
