番茄碱的分离纯化和抑菌活性的研究

摘    要:目的:研究番茄碱的分离纯化工艺及其对大肠杆菌的抑菌活性。方法采用乙醇提取青番茄中的番茄苷,水解后得番茄碱,经 HPd-100A大孔吸附树脂分离纯化,采用高效液相-质谱/质谱(HPLC-MS/MS)检测番茄碱含量;采用琼脂扩散滤纸片法研究番茄碱对大肠杆菌的抑制活性,确定番茄碱的最低抑菌浓度,考察不同 pH值、温度、作用时间对抑菌活性的影响。结果番茄碱粗品经上述工艺纯化后,含量达到39.05%;番茄碱的最低抑菌浓度为113.2 mg/mL,在 pH为7、作用时间为12 h、温度为90℃条件下抑菌活性最好。结论 HPd-100A大孔吸附树脂对番茄碱具有较好的分离效果,一定浓度下的番茄碱可抑制大肠杆菌生长。

Abstract:Objective To study the purification process of tomatine and its antibacterial activity on Escherichia coli.Methods Used the ethanol to extract tomato glycosides from green tomatoes,hydrolyzed to tomatine,after further purified by HPd-100A macroporous resin,then detected content of tomatine by HPLC/MS/MS;Agar diffusion filter paper method was used to study the inhibitory activity of tomatine to Escherichia coli,minimal inhibitory concentration (MIC)tomato glycosides on Escherichia coli were determined,investigated the influence of different pH value,temperature,reaction time on their antibacterial activity.Results After the above process purificated,the content of raw tomatine reached to 39.05%;the MIC of tomatine was 113.2 mg/mL,when temperature was 90℃,pH was 7,action time was 12 hours, antibacterial activity was better.Conclusion HPd-100 macroporous resin for tomatine has better separation effect,under a certain concentration of tomatine can inhibit growth of Escherichia coli.
