泽泻汤加味方对高盐致高血压大鼠肾损害的预防作用

摘    要:目的 探讨泽泻汤加味方对高血压大鼠肾损害的预防作用及其机制.方法 给Wistar大鼠高盐饮食22周造模,将40只高血压大鼠随机分为模型组、缬沙坦组、泽泻汤加味方高、中剂量组(高、中剂量组),每组10只,另设正常组10只.从第23周起,模型组给予生理盐水,缬沙坦组予缬沙坦溶液14.4mg/(kg·d),泽泻汤加味方高、中剂量分别予泽泻汤加味方混悬液16.2g/(kg·d)、10.8g/(kg·d),正常组正常饲养.干预8周后,对其肾脏行病理诊断;观察各组大鼠尿蛋白(ALB)、尿β2-微球蛋白(β2 -MG)、肾组织肾素(RN)活性及血管紧张素Ⅱ(AngⅡ)的变化.结果 与模型组比较,高剂量组大鼠24h尿ALB明显降低(P<0.01);中剂量组尿β2-MG降低(P<0.05).免疫组化结果显示,AngⅡ在模型组和药物治疗组肾脏的肾小球均为强阳性,而正常组为弱阳性.与正常组比较,各组大鼠肾组织RN活性明显升高(P<0.01),中剂量组AngⅡ含量明显升高(P<0.01).结论 泽泻汤加味方可改善高盐致高血压大鼠肾脏的结构和功能,其机制与上调肾脏RN活性和AngⅡ水平有关,从而起到预防肾损害的作用.

Effect of Modified Zexie Decoction on Prevention of Kidney Injuries of Rats with Hypertension induced by High-Salt Diet

Abstract:Objective To discuss the effect of Modified Zexie Decoction (Decoction of Rhizoma Alismatis plus other Chinese herbal medicines) on the prevention of kidney injuries of rats with hypertension induced by high-salt diet. Methods Wistar rats were fed with high salt diet for 22 weeks to establish hypertension models. The successfully-made model rats were divided into model group, valsartan group, and Modified Zexie Decoction high-dose group and mid-dose group with 10 rats in each group. From the 23rd week, the model group was given normal saline, the valsartan group given valsartan 14.4mg/(kg·d), Modified Zexie Decoction high-dose group and mid-dose group given suspension of the decoction 16.2g/(kg·d) and 10.8g/(kg·d) respectively. Additional 10 normal rats were as normal group given normal diet and water. After 8 weeks, the pathologic diagnosis of kidney was carried out and the changes of urine albumin (ALB), urine β2-microglobulin (β2-MG), the activity of renin (RN) of renal issues, and angiotensin Ⅱ(AngⅡ) were observed. Results Compared with the model group, the 24h-ALB of Modified Zexie Decoction high-dose group was significantly decreased (P<0.01), and the urine β2-MG of Modified Zexie Decoction mid-dose group was significantly decreased (P<0.05). The result of immunohistochemistry test showed that the AngⅡ was strongly positive in glomeruli of the model group and medication groups, but it was weakly positive in glomeruli of the normal group. Compared with normal group, the activity of RN of other groups was significantly increased (P<0.01), and the AngⅡ level of Modified Zexie Decoction mid-dose group was significantly increased (P<0.01). Conclusion The Modified Zexie Decoction is effective in improving the construction and function of model rats, and this may be concerned with the increasing of RN activity and AngⅡ level of renal tissues, thus it prevents the kidney injuries.
