益气补肾方对胃癌细胞侵袭转移能力的影响

目的 探讨益气补肾方对胃癌细胞SGC-7901侵袭转移能力的影响.方法 将胃癌SGC-7901细胞接种于RPMI-1640培养液中,益气补肾方水溶液配制成1、2、4、8、16、32、64mg/ml 7个终浓度,设肿瘤细胞为空白对照组,在96孔板内每个剂量6个平行孔,重复3次,37℃、5％CO2培养箱孵育48h.采用MTT法检测不同浓度益气补肾方对SGC-7901细胞增殖的抑制率；采用RT-PCR法检测其对SGC-7901细胞基质金属蛋白酶-2(MMP-2)和基质金属蛋白酶抑制酶-2(TIMP-2)的基因表达活性的影响. 结果 不同浓度的益气补肾方对SGC-7901细胞都有一定的抑制作用,以16mg/ml和32mg/ml浓度抑瘤作用较好,抑制率分别为53.17％和56.47％；MMP-2 mRNA在32mg/ml组同空白对照比较,差异有统计学意义(P＜0.05),并且16mg/ml组也能下调MMP-2 mRNA的表达,但差异无统计学意义(P＞0.05). 结论 益气补肾方能明显抑制胃癌细胞SGC-7901侵袭转移能力,且以32mg/ml浓度最好.

Influence of Yiqi Bushen Formula on the Ability of Invasion and Metastasis of Gastric Cancer Cells

Objective To discuss the influence of Yiqi Bushen Formula (Formula for replenishing qi and tonifying the kidney) on the ability of invasion and metastasis of gastric cancer cells SGC-7901. Methods The gastric cancer cells SGC-7901 were inoculated into the RPMI-1640 culture solution. Yiqi Bushen Formula decoction was prepared to 7 final concentrations as 1, 2, 4, 8, 16, 32, and 64mg/ml. The cancer cells, taken as the blank control group, were put into the 96-hole plate. Each dose of the Formula decoction was applied to the 6 parallel holes of the plate. The procedure was repeated for 3 times. Cells were incubated in the incubator at 37℃ and 5%CO2 for 48h. The inhibition rate of proliferation of SGC-7901 cells by the different concentrations of Yiqi Bushen Formula was detected with the MTT method. The activity of gene expressions of matrix metalloproteinase-2 (MMP-2) and its inhibitory enzyme-2 (TIMP-2) of SGC-7901 cells were measured with the RT-PCR method. Results The Formula decoction of different concentrations had certain inhibiting action on SGC-7901 cells, but the concentrations 16mg/ml and 32mg/ml functioned better, the inhibition rate being 53.17% and 56.47% respectively. Compared with blank control group, the difference was statistically significant in the MMP-2 of the 32mg/ml group (P<0.05), and the MMP-2 mRNA expression of the 16mg/ml group was also down-regulated, but the difference was not statistically significant (P>0.05). Conclusion Yiqi Bushen Formula is effective in inhibiting the ability of invasion and metastasis of gastric cancer cells SGC-7901, and the result of concentration 32mg/ml was the best.
