蛋白芯片法在不孕不育抗体检测中的应用

摘    要:目的评价不孕不育蛋白芯片的临床应用价值。方法用不孕不育蛋白芯片对430份临床标本进行抗精子抗体(ASA)、抗子宫内膜抗体(AEA)、抗心磷脂抗体(ACA)、抗卵巢抗体(AOA)、抗人绒毛膜促性腺激素(HCG)抗体检测,并与化学发光法(ECLIA)检测结果进行对比。结果蛋白芯片法与ECLIA法的ASA、ACA、AEA和HCG的检测结果具有较好的一致性,但在AOA的检测结果上差异有统计学意义(χ^2=4.5,P〈0.05)。其中ASA的阳性符合率为91.8%,阴性符合率为98.8%;ACA的阳性符合率为92.1%,阴性符合率为96.0%;AEA的阳性符合率为87.5%,阴性符合率为95.8%;AOA的阳性符合率为79.1%,阴性符合率为98.9%;抗HCG的阳性符合率为90.9%,阴性符合率为98.4%。结论从整体检测结果上来看,不孕不育抗体蛋白芯片联检试剂盒具有操作简便、快速和特异性强等优点,值得在临床上推广使用,但其AOA检测的灵敏度还有待于提高。

Application of Protein Chip Technology in The Antibody Test of Infertility and Sterility

Abstract:[ Objective ] To evaluate the clinical value of protein chips technology in the antibody test of the infertility and sterility. [ Methods] A protein chip method was employed to detect anti-sperm antibody( ASA), anti-endometrial antibody( AEA), anti-cardiolipin antibody (ACA) , anti-ovarian antibody( AOA) and anti-human chorionic gonadotropin (HCG) antibody of 430 clinical specimens, and the results were compared with enzymatic chemiluminescence (ECLIA). [ Results ] The detecting results of ASA. , ACA, AEA and HCG with protein chip were consistent with ECLIA results ( P 〉 0.05 ) , but the results of AEA were significant different ( χ^2 = 4.5, P 〈 0.05 ). The coincidence rate of positive results of ASA, ACA, AEA, AOA and HCG was 91.8%, 92.1%, 87. 5%, 79.1% and 90.9% respectively, while the coincidence rate of negative restdts was 98.8%, 96.0%. 95.8%, 98.9% and 98.4% respectively. [ Condusion] Infertility and sterility antibodies detection by protein chip method is simple, rapid and specific, and it is worthy of clinic application, but the sensitivity of AOA detection remains to be improved.
