冻融人早孕蜕膜单层细胞对小鼠胚胎发育的影响

摘    要:目的 探讨冻融的人早孕蜕膜单层细胞对小鼠胚胎发育的影响。方法 蜕膜单层细胞解冻后接种获得冻融蜕膜单层细胞;观察昆明白小鼠单细胞受精卵与传代的、冻融的人早孕蜕膜单层细胞共培养 (A、B组)及仅用培养液培养(C组)的胚胎发育情况。分析A、B组与C组各期鼠胚的发育情况及 8 细胞期A类胚胎形成情况。结果 A、B、C组的2 细胞期形成率差异无显著性,A组在 4 细胞形成率上高于B、C组。A、B组 8 细胞期、桑椹期、早期囊胚形成率和囊胚孵出率高于C组(P<0 05)。而A、B组之间差异无显著性。A、B组在 8 细胞阶段A、B类胚胎形成率与C组相比,差异有显著性(P<0 05)。同样A、B组之间差异无显著性。共培养 20d的培养液经检查,细菌及真菌均阴性。结论 冻融的人蜕膜单层细胞与传代细胞一样在小鼠胚胎的发育中起到了促进作用,该结果说明了人胚胎与其冻融的自体子宫增殖晚期-分泌早期的内膜单层细胞共培养的可行性。

Abstract:Objective To evaluate the effects of human frozen thawed decidua monolayer cells on mouse embryo development in a co culture system. Methods Kunming mouse 1 cell embryos obtained by controlled ovarian superovulations were cultured with fresh or frozen thawed human decidua monolayer cells (marked in group A and B, respectively), or cultured with conventional media (group C). Results The formation rates of 8 cell embryos, morulae, early blastocysts and expanded blastocysts were significantly lower in group C, whereas that was no difference between groups A and B. The formation rate of high quality embryos in groups A and B was significantly higher than that in group C. Pathogen was examined in culture medium, and bacteria and fungus were not found by 20 days. Conclusion The frozen thawed human decidua monolayer cells as a kind of feeder can improve the mouse embryo development. Contamination can be avoided by strictly aseptic operation.
