原发性骨质疏松骨组织c-fos、c-jun和p53基因的表达

摘    要:目的 研究原发性骨质疏松时骨组织中原癌基因c-fos、c-jun和抑癌基因p53的表达情况,以探讨c-fos、c-jun和p53与原发性骨质疏松症的关系及其在骨质疏松中所起的作用. 方法 采用人的新鲜松质骨作为实验标本(骨质疏松组和对照组),分别用兔抗人c-fos、c-jun多克隆抗体和鼠抗人p53单克隆抗体通过免疫组化方法来检测标本中c-fos、c-jun和p53基因的蛋白产物,了解c-fos、c-jun、p53基因的表达情况。结果 骨质疏松组的骨细胞和成骨细胞中c-fos、c-jun、p53基因的阳性表达率均为100%,与对照组相比显著增高,P<0.01。结论 原发性骨质疏松时骨组织中c-fos、c-jun、p53基因的表达增高,提示癌基因c-fos、c-jun和抑癌基因p53与原发性骨质疏松的关系密切,原发性骨质疏松时c-fos、c-jun、p53表达增高与骨细胞和成骨细胞的凋亡有关。

Abstract:Objective To study the expression of proto - oncogene c - fos , c - jun and antioncogene p53 in order to understand the relationship between these genes and osteoporosis, and their effects on primary osteoporosis. Methods The fresh cancellous bones of the patients who had undergone orthopedics operations were divided into two groups (osteoporosis and control) . The protein products of c - fos, c - jun and p53 in the samples were detected with the their antibodies respectively by immunohistochemistry method. Results It was found that the expressions of c - fos, c - jun and p53 all increased in osteocytes and osteoblasts in the osteoporosis group, being significantly different from the control group, P<0.01 . Conclusions The expressions of c - fos , c - jun and p53 increased in primary osteoprosis. It suggested that proto - oncogene c - fos , c - jun and antioncogene p53 might play an important role on the mechanism of primary osteoporosis. The increased expressions of c - fos, c - jun and p53 may be concerned with the apoptosis of osteocytes and osteoblasts in primary osteoporosis.
