| 摘 要： | 目的：用不同剂量的ＶＰ－１６诱导急性早幼粒白血病细胞程序化死亡（ＰＣＤ）。方法：通过细胞培养技术获取ＰＣＤ细胞，以ＤＮＡ琼脂糖凝胶电泳确定细胞ＰＣＤ的发生，苔盼蓝拒染法检测细胞活力。结果：不同浓度的ＶＰ－１６在试验条件下于４ｈ内均能诱导ＨＬ－６０细胞发生ＰＣＤ。结论：本实验建立的研究细胞ＰＣＤ的方法可靠，ＶＰ－１６具有良好的诱导细胞产生ＰＣＤ的作用

| Abstract: | OBJECTIVE:This study was aimed to observe the etoposide (VP16)induced programmed cell death (PCD) of the promyelocytic leukemic HL60 cell.METHODS:HL60 cells were cultured in RPMI 1640 medium containing 20% calf serum.For experiments,the medium was supplemented with VP16.The PCD cells obtained from the culture cells were conformed by DNA electrophoresis and viability of cells with trypan blue test.RESULT:PCD of HL60 cells occurs in different concentration of VP16 within 4 hours.CONCLUSION:Our method is suitable for studying PCD and VP16 has a function to induce PCD. |
