甲基磺酸乙酯化学诱变川芎的方法研究

摘    要:目的:通过甲基磺酸乙酯(EMS)诱导川芎材料突变,筛选最佳诱变条件为川芎新品种的培育研究提供方法学基础。方法:本实验在组织培养基础上,使用多浓度(0.1%、0.2%、0.5%、1.0%)的甲基磺酸乙酯(EMS)对川芎茎节进行不同时间段(0.5 h、1 h、2 h、5 h、8 h、12 h)的化学诱变处理,观察无菌试管苗的生长速度、发育形态,比较优化EMS诱变川芎无菌苗的最佳条件。结果:经0.1%EMS、1%EMS处理1 h、5 h的川芎茎节较对照粗壮且生长快速。结论:0.1%EMS、1%EMS处理1h、5h为川芎茎节的最佳诱导条件。

Abstract:Objective:To induce the mutant of chuangxiong by chemical mutagenesis.creening mutagenesis optimum conditions for the cultivation as new varieties of Ligusticum chuanxiong Hort to provide methodological basis.Methods:To select the survived jointed stems which has been treated by EMS with different concentration (0.1%,0.2 %,0.5 %,1%) and time (0.5 h,1 h,2 h,5 h,8 h,12 h) basis on the tissue culture,observation of sterile plantlet growth speed,morphological development.Comparison and optimization the best conditions for EMS mutation of sterile seedlings of Ligusticum chuanxiong.Results:The jointed stems treated with 0.1% EMS and 0.5 % EMS 1 h,5 h grows faster and robuster.Conclusion:The jointed stems treated with 0.1% EMS and 0.5 % EMS 1 h,5 h are the optimal inducing condition.
