解偶联蛋白2在蜕膜巨噬细胞活性调控中的作用及与复发性流产的关系

摘    要:目的探讨蜕膜巨噬细胞解偶联蛋白2(UCP2)表达、细胞内活性氧(ROS)、吞噬活性之间的关系及其在原因不明复发性流产发病中的作用。方法采用免疫磁珠法分离11例原因不明复发性流产患者和12例正常早孕妇女蜕膜巨噬细胞,流式细胞技术检测两组蜕膜巨噬细胞UCP2的表达、细胞内ROS水平;同时检测巨噬细胞吞噬活性。结果蜕膜巨噬细胞UCP2表达与细胞内ROS水平负相关(r=-0.572,P0.01),与巨噬细胞吞噬活性负相关(r=-0.462,P0.05)。与正常早孕妇女比较,原因不明复发性流产患者蜕膜巨噬细胞UCP2表达明显下降,细胞内ROS水平升高,其吞噬活性增强。结论UCP2通过控制巨噬细胞内ROS水平,调节细胞活性,在维持正常妊娠免疫耐受和原因不明复发性流产发病中起重要作用。

Abstract:Objective: To investigate the relationship between the expression of uncoupling protein 2 (UCP2), reactive oxygen species (ROS) level and phagocytic activity of decidual macrophages and the occurrence of unexplained recurrent spontaneous abortion (URSA). Methods: Decidual tissues were collected from 12 women with selective termination of normal early pregnancy and 11 women with URSA. Magnetic cell sorting (MACS) was used to isolate the monoeyted macrophages in decidua. The expression of UCP2 and ROS level in decidual macrophages was detected with flow cytometry. Neutral red stain was used to check the phagocytic activity of macrophages. Results: The expression of UCP2 in macrophages was negatively correlated to ROS level (r=-0. 572, P〈0.01) and phagocytic activity (r=-0.462, P%0.05). Compared to that in control group, the expression of UCP2 in decidual macrophages of women with URSA was decreased significantly (136.45±25.41 vs. 201.25±30.96, P〈0.01), while the ROS level and phagocytic activity were higher. Conclusions: UCP2 may play an important role in the regulation of macrophage activity and contribute to spontaneous abortion.
