化疗栓塞对大鼠Walker-256肝肿瘤移植模型微血管密度、VEGF和bFGF表达的影响

摘    要:目的:用大鼠Walker-256肝肿瘤移植模型研究化学栓塞对肿瘤微血管密度(MVD)、血管内皮细胞生长因子(VEGF)和成纤维细胞生长因子(bFGF)表达的影响。方法:60只大鼠肝肿瘤模型在种植后第14天随机分成3组,分别经肝动脉注入超液态碘油0.5 mL(A组)、超液态碘油0.5 mL加5-Fu 20 mg•kg-1(B组)和生理盐水0.5 mL (对照组)。于术后第7天取肝脏标本,免疫组化法检测肿瘤组织MVD、VEGF和bFGF的表达,同时分析VEGF表达强度与MVD的相关性。结果:各组MVD 差异无显著性(P>0.05);A组和B组瘤内VEGF阳性率和平均染色强度明显高于对照组(P<0.05, P<0.01),而A组与B组之间差异无显著性(P>0.05)。VEGF表达强度与MVD呈显著正相关(r=0.451,P<0.01)。bFGF阳性率和平均染色强度各组差异无显著性(P>0.05)。结论:化疗药物栓塞后残存瘤组织具有丰富的血供,提高了残存肿瘤组织VEGF的表达,VEGF和MVD可能在术后肿瘤血供的重建中起重要作用。

Abstract:Objective To study the expression of microvessel density (MVD), vascular endothelial growth factor (VEGF), and basic fibroblast growth factor (bFGF) in the rat model with Walker 256 hepatoma after chemoembolization. Methods Sixty rats with Walker 256 hepatoma were divided into 3 groups randomly at the 14th day after the tumors were implanted. Lipiodol 0 5 mL (TAIE group), lipiodol 0 5 mL plus 5 Fu 20 mg·kg -1 (TACE group) and routine saline 0 5 mL (control group) were injected in different groups via hepatic artery to embolize tumor of rats. After 7 days of treatment, the expressions of MVD, VEGF, and bFGF in tumor tissues were examined with immunohistochemical technique. The correlation of VEGF expression intensity and MVD was analyzed.Results There was no significant difference of MVD between any two of the three groups ( P >0 05). The positive rate and staining intensity of VEGF were significant higher in TAIE group and TACE group than those in control group ( P <0 05, P <0 01), but there were no significant difference between TAIE and TACE group( P >0 05). The staining intensity of VEGF was significantly correlative to the MVD in tumor tissue ( r=0 451, P <0 01) . There were no significant differences of the positive rate and staining intensity of bFGF between any two of the three groups ( P >0 05). Conclusion Survival tumor tissue was supplied with rich vascularity after TACE. The expression of VEGF was enhanded after TACE. VEGF and MVD may play an important role in reestablishment of bood supply for the tumor after TACE.
