MMP-2和TIMP-2在非小细胞肺癌中的表达及其临床意义

摘    要:目的 :探讨基质金属蛋白酶 (matrixmetalloproteinases ,MMPs)MMP 2和组织金属蛋白酶抑制物(tissueinhibitorofmetalloproteinases ,TIMPs)TIMP 2在非小细胞肺癌 (NSCLC)中的表达及其与肿瘤血管形成和淋巴结转移的关系。方法 :用免疫组化 (S P法 )染色技术对 95例NSCLC石腊组织标本的MMP 2、TIMP 2和肿瘤内微血管密度 (IMVD)进行检测和分析。结果 :MMP 2和TIMP 2在肺癌细胞中阳性表达率明显高于癌旁的正常肺组织 ,P <0 0 1,而且MMP 2在间质的纤维细胞中亦大量表达。MMP 2阳性表达癌组织中的IMVD高于阴性表达癌组织中的IMVD ,P <0 0 5 ;TIMP阳性表达癌组织中的IMVD则低于阴性表达癌组织中的IMVD ,P <0 0 5。MMP 2表达与淋巴结转移呈正相关 ,而TIMP 2与其呈负相关 ,P <0 0 5。MMP 2表达随着肿瘤病理分级和TNM分期增高而有增高趋势 ,而TIMP 2的阳性表达情况则有下降趋势。结论 :MMP 2和TIMP 2可能均参与了NSCLC新血管生成的调节 ,二者表达失衡促进肿瘤的侵袭和转移 ,MMP 2和TIMP 2的检测可作为判断NSCLC转移及预后的指标

Abstract:Objectives To investigate the expression and distribution of MMP 2 and TIMP 2 in human non small cell lung cancer (NSCLC),and to evaluate the relationship between their expression and tumor angiogenesis and lymph node metastasis.Methods The expression of MMP 2 and TIMP 2 in 95 patients with NSCLC were examined by immunohistochemical methods (S P),and their relationship to some clinicopathological parameters was analyzed.Results The positive rates of MMP 2 and TIMP 2 in NSCLC were much higher than those in the normal lung tissues nearby, P <0 01.The positive expression of MMP 2 and TIMP 2 was associated with intratumor microvessel density(IMVD),pathological grades and lymph node metastases of NSCLC, P <0 05.Conclusions The expression and distribution suggested that MMP 2 and TIMP 2 might play an important role in tumor angiogenesis and lymph metastasis of human NSCLC,and detection of MMP 2 and TIMP 2 may be valuable for prediction of human NSCLC metastasis and prognosis.
