摘    要:目的:建立以固相萃取-HPLC法测定人血浆中双氯芬酸钠浓度的方法。方法:色谱柱为ODS-2,流动相为磷酸盐缓冲液(pH6.5)-甲醇(40∶60),检测波长为280nm,流速为1.0mL.min-1。结果:双氯芬酸钠血浆浓度在16.2~2 024.0ng.mL-1范围内与峰面积积分值线性关系良好(r=0.998 9),方法回收率在90%~110%之间,萃取回收率大于90%;日内、日间相对标准差小于5%。结论:本方法操作简便,结果准确可靠,可用于双氯芬酸钠药动学研究。

Abstract:OBJECTIVE: To establish an SPE(solid phase extraction)-HPLC method for the determination of diclofenac sodium in human plasma.METHODS: The separation of diclofenac sodium was performed on ODS-2 column with a detection wavelength of 280nm.The mobile phase was composed of phosphate buffer(pH6.5)-methanol(40∶ 60) at a flow rate of 1.0mL· min-1.RESULTS: The linear range of diclofenac sodium was 16.2~ 2 024.0ng· mL-1(r=0.998 9).The methodological recovery ranged between 90% to 110% and the extraction recovery was above 90%.Both intraday RSD and inter-day RSD were less than 5%.CONCLUSION: The method is simple,accurate and reliable,and suitable for the pharmacokinetic study of diclofenac sodium.
