阿魏酸对血管平滑肌细胞增殖的抑制作用研究

目的研究阿魏酸对血管平滑机细胞增殖的影响。方法采用贴块法培养人脐动脉血管平滑肌细胞(VSMC);台盼兰染色法检测药物对细胞的毒性;以四甲基偶氮唑蓝(MTT)法观察药物对血清诱导的VSMC增殖及生长的影响;以Bradford法检测细胞总蛋白含量变化;用5溴-2脱氧尿苷(BrdU)免疫酶联吸附法观察对DNA合成的影响。结果阿魏酸可逆性的延迟细胞对数生长期,影响细胞生长;有效抑制细胞的增殖活性,当阿魏酸终浓度为100μmol/L时,对细胞增殖的抑制率达到61.32%;引起细胞总蛋白的合成及BrdU的DNA掺入量下降,影响细胞周期。结论阿魏酸有效抑制血清诱导的VSMCs增殖,对以VSMCs异常增殖为靶标的动脉粥样硬化等心脑血管疾病的预防和治疗具有积极作用。

INHIBITION OF FERULIC ACID ON VASCULAR SMOOTH MUSCLE CELLS PROLIFERATION

Objective To investigate the inhibition of ferulic acid(FA) on vascular smooth muscle cells(VSMCs) proliferation.Method Human umbilical VSMCs were obtained from umbilical artery by the attached explant method.The cell cytotoxicity of FA was measured by trypan blue exclusion test,and the effect of FA on serum-induced proliferation of VSMCs was evaluated by MTT assay.The variance of total protein was determined by Bradford method,and DNA synthesis was investigated by human BrdU ELISA.Result FA reversibly delayed the logarithm growth times of VSMC.At concentration of 100 μmol/L,FA inhibited cell proliferation by 61.32% without significant cytotoxicity.Moreover,FA inhibited the cell protein and DNA synthesis in dose-dependent manner.Conclusion Ferulic acid might contribute to the prevention and treatment of atherosclerosis through inhibiting SMC proliferation.
