鹿石合剂对缺氧缺血脑损伤新生大鼠海马BDNF、NGF、NSE免疫表达的影响

摘    要:目的探讨鹿石合剂对缺氧缺血脑损伤新生大鼠保护作用的机制。方法建立缺氧缺血新生大鼠动物模型,用HE染色及免疫组织化学方法,观测药物干预后大鼠海马CA1区锥体细胞形态学变化及对脑源性神经营养因子(BDNF)、神经生长因子(NGF)和神经元烯醇化酶(NSE)免疫阳性神经元的影响。结果鹿石合剂高剂量及“洛斯宝”干预后,细胞形态接近空白组;对于BDNF及NGF,与模型组及鹿石合剂低剂量组比较,P〈0.01;对于NSE,免疫反应接近空白组。结论鹿石合剂可上调大鼠海马CA1区锥体细胞BDNF、NGF及NSE的免疫表达水平,促进神经元的修复及神经功能的重塑。

Abstract:Objective To explore the protective effect of Lushi misture on hypoxic-ischemic brain damage neonatal rats. Methods The rat models were made and then interfered with Lushi misture, and vasobral as control. The changes of pyramidal neurone's morphology and BDNF, NGF and NSE's immune positive neurons in hippocampus CA1 section were observed by HE dyeing and immunohistochemistry methods. Results The cell shapes of high dose group of Lushi misture and Vasobral group approach to normal group, the levels of BDNF and NGF were higher than that of low dose group and non-drug group (P<0.01), and the levels of NSE approach to the blank group. Conclusion Lushi misture can up-regulate the expression levels of BDNF, NGF and NSE in hippocampus CA1 section pyramidal neurons of hypoxic-ischemic brain damage neonatal rats, thus encourage the reparation of the hypoxic-ischemic neurons and neurofunctional remodeling.
