雾化吸入三氧化二砷对哮喘豚鼠气道嗜酸粒细胞影响的研究

摘    要:目的 通过观察雾化吸入三氧化二砷(As2O3)对哮喘豚鼠气道嗜酸粒细胞的病理改变,探讨As2O3剂雾化吸入的平喘治疗的可行性。方法 采用鸡卵蛋白(OVA)复制哮喘豚鼠模型,分组给予不同剂量As2O3喷射雾化吸入;同时设生理盐水雾化吸入组、大剂量的As2O3腹腔注射组对照,均用药7d后比较支气管粘膜下层的嗜酸粒细胞(EOS)的情况。结果 小剂量的As2O3(1.0和2.0mg/kg·d-1)雾化吸入组支气管粘膜下层的EOS明显减少,与哮喘组比较差异有高度显著性(P<0.01);与较大剂量的As2O3(5.0mg/kg·d-1)腹腔注射组比较差异无显著性(P>0.05)。结论 小剂量的As2O3雾化吸入能够显著减少哮喘豚鼠气道中的EOS,从而减轻哮喘的气道炎症,短期使用可以相对安全而有效地达到治疗哮喘的目的。

Abstract:Objective To study the feasibility of aerosol inhalation of arsenic trioxide(As2O2) in treatment of asthma guinea pigs by observing the pathological changes of the eosinophils (BOS) in bronchial mucosa in the guinea pigs after aerosol inhalation of ( As2O3). Methods Thirty - six guinea pigs asthma models were reproduced by using OVA. Then they were randomly allocated to three groups: group A (asthma control group, n =6), group B (As2O3 management group, and subdivided into groups B1, B2 and B3 of which inhalation As2O3 0.5, 1.0 and 2.0 mg/kg·d-1, respectively, as well as group B4 of which peritoneal injection As2O3 5. 0mg/kg·d-1), and group C (the normal control group). Both groups A and C were inhalated in normal saline. Seven days after the management, all the EOS condition in the lower bronchial mucosa were analyzed and compared with each other. Results The number of EOS reduced significantly in groups B2 and B3, in comparison to those in group A, the differences were significntly different ( P <0.01); but there were no difference in comparison to those in group B4( P >0.05). Conclusion Aerosol inhalation of small dosage of As2O3 could significantly reduce the number of bronchial EOS in asthma guinea pigs so that the inflammation sympton could be remittent. Short - term inhalation of As2O3 could be safe and effective in treatment of asthma.
