TLR4及TAK1表达与急性脑梗死炎性反应机制相关性研究

摘    要:目的:探讨急性脑梗死患者外周血中Toll样受体4(TLR4)及转化生长因子激活激酶1(TAK1)表达的变化及其与炎性反应机制的相关性。方法:对50例急性脑梗死患者进行临床特征分析、外周血白细胞计数、CRP水平、NIHSS评分、脑梗死灶容积计算及血清TLR4及TAK1的含量检测,分析梗死灶容积与上述指标的相关性。结果:急性脑梗死患者血糖水平、CRP水平、白细胞计数、NIHSS评分、梗死灶容积均较健康对照组高,差异比较均有统计学意义(P<0.05);急性脑梗死整个发病过程中血清中TLR4与TAK1含量均较健康对照组高,以疾病发病高峰期最明显;脑梗死灶容积与糖尿病史、CRP水平、白细胞计数及NIHSS评分有相关性,其中TLR4与脑梗死灶容积有独立相关性。结论:TLR4及TAK1可能主要是通过介导炎性反应在脑梗死中发挥作用,其中TLR4与脑梗死容积有独立相关性,可作为脑梗死的治疗靶点。

Abstract:Objective:To explore expressions of Toll-like receptor 4(TLR4)and TGF-activated kinase 1(TAK1)in acute ischemic stroke and their correlation with mechanisms of inflammatory reaction.Methods:Clinical characters of 50 cases with acute ischemic stroke were analyzed.The counts of WBC in peripheralc,ontent of CRP、TLR4 and TAK1 in serum were detecteds,coring of NIHSSv,olumn of infarct were calculated.Finally,correlation between volumn of infarct and the above indices was analyzed.Results:The blood glucose levelc、ontent of CRPc、ounts of WBCs、coring of NIHSS and vol-ume of infarct were higher in acute ischemic stroke group than those in control group;content of TLR4 and TAK1 were in-creased in acute ischemic stroke groupv;olume of infarct was associated with count of WBC,CRPs,coring of NIHSS and diabetes history;TLR4 was independently relevance to volume of infarct.Conclusion:TLR4 and TAK1 may play an im-portant role in developing of acute ischemic stroke by enhancing inflammation.TLR4 may be considered as a potential therapeutic target for acute ischemic stroke.
