摘    要:目的 建立阿尔茨海默病(AD)双转基因鼠,为进一步研究AD发病机制提供较为理想的实验动物.方法 将人载脂蛋白E4转基因鼠和突变APP转基因鼠杂交.结果 经PCR初筛,对阳性小鼠基因组DNA作进一步的Southern杂交鉴定,获得2只双转基因小鼠,之后传代建系.结论 为进一步研究多个基因对AD的致病作用提供较为理想的实验动物.

Abstract:Objective To establish animal model of Alzheimer's disease(AD) in transgenic mice and investigate the mechanism of AD.Methods To establish AD bi-transgenic mice by ApoE4 and APP transgenic mice intercrossing.The PCR and Southern blot hybridization techniques were used for identification of transgenic mice.Results Two ApoE4/APP transgenic mice were born and young mice were bred.Conclusions ApoE4 and APP transgenic mice is a good AD animal model.
