摘    要:目的 探讨表皮生长因子受体(EGFR)基因对胰腺癌细胞放疗敏感性的增强作用.方法 构建针对EGFR基因序列特异性shRNA的表达载体,用脂质体转染胰腺癌PANC-1细胞,G418筛选建立稳定克隆,采用RT-PCR、Western印迹检测EGFR基因的抑制情况.采用不同剂量X线照射瘤细胞后,用MTT法检测细胞凋亡率,集落形成实验检测细胞集落形成能力,观察胰腺癌细胞对放疗的敏感性.结果 靶向EGFR的序列特异性shRNA可明显抑制EGFR基因的表达,EGFR mRNA和蛋白的抑制率分别为76.3%和71.4%;细胞凋亡增加(P<0.01),克隆形成率减少(P<0.01).结论 体外实验证实靶向EGFR的序列特异性shRNA可明显提高胰腺癌的放疗敏感性.
