乳腺癌患者外周血ALDH1~(high)CD44~+CD24~(-/low)细胞含量变化及其临床意义

摘    要:目的:检测乳腺癌患者外周血ALDH1highCD44+CD24-/low细胞含量的变化,探讨其与乳腺癌发生的关系及临床意义。方法:观察对象为46例Ⅰ期、Ⅱ期、Ⅲ期和Ⅳ期乳腺癌患者和12名健康女性志愿者(对照组)。采用流式细胞仪检测、分析各组外周血中ALDH1high细胞和ALDH1highCD44+CD24-/low细胞含量及其变化情况。结果:①所有样本均能检出ALDH1high细胞,各组间均数比较差异无统计学意义(P>0.05);②Ⅳ期乳腺癌组的外周血ALDH1highCD44+CD24-/low细胞含量与其他各组比较差异均有统计学意义(P<0.05),而其他各组间比较差异无统计学意义(P>0.05);③Ⅳ期乳腺癌患者原发癌灶免疫组化结果各不相同,但均可在外周血中检出ALDH1highCD44+CD24-/low细胞。结论:①各组中的ALDH1high细胞含量无显著差异,说明ALDH1具有普遍干细胞性质;②ALDH1highCD44+CD24-/low细胞含量与乳腺癌的发生有明显相关性。③ALDH1highCD44+CD24-/low细胞与乳腺癌的免疫分型无相关性。

Change and clinical significance of ALDH1highCD44+CD24-/low cells content in peripheral blood of patients with breast cancer

Abstract:Objective:To detect the change of ALDH1highCD44+CD24-/low cells content in peripheral blood of patients with breast cancer,explore its relationship with the occurrence of breast cancer and its clinical significance.Methods:Forty-six patients with breast cancer of stage Ⅰ,Ⅱ,Ⅲ,and Ⅳ,and twelve healthy women(control group) were selected as observation groups;flow cytometry was used to detect and analyze the content of ALDH1highCD44+CD24-/low cells in peripheral blood and the change.Results:ALDH1high cells could be detected in all the samples,there was no statistically significant difference in the mean among different groups(P>0.05).There was statistically significant difference in the content of ALDH1highCD44+CD24-/low cells in peripheral blood between breast cancer group of stage Ⅳ and the other groups(P<0.05),but there was no statistically significant difference among the other groups(P>0.05).Among the patients with breast cancer of stage Ⅳ,the immunohistochemical results of primary cancer varied,but ALDH1highCD44+CD24-/low cells could be detected in every sample.Conclusion:The content of ALDH1high cells in different groups had no statistically significant difference,which indicates that ALDH1 is a universal stem cell marker.The content of ALDH1highCD44+CD24-/low cells is significantly correlated with the occurrence of breast cancer.There is no correlation between ALDH1highCD44+CD24-/low cells and immunophenotype of breast cancer.
