摘    要:目的探讨移植转染pDsVEGF165Red1—N1质粒的神经干细胞能否促进脑外伤大鼠神经功能的恢复。方法利用阳离子脂质体转染大鼠神经干细胞,在脑立体定向仪引导下分别将PBS(A组)、神经干细胞(B组)、转基因神经干细胞(C组)移植到脑外伤大鼠局部损伤灶边缘,通过NSS评分评价移植后大鼠神经功能的变化。结果转基因细胞移植后可以在一定时间内表达VEGF165,C组大鼠NSS评分从移植后第3天开始明显低于A组(P<0.05),而B组大鼠NSS评分则从移植后第7天开始明显低于A组(P<0.05)。结论转基因神经干细胞移植后可以分泌VEGF165促进脑外伤大鼠神经功能的恢复。

Abstract:Objective To explore the protection of neural stem cells transfected with pDsVEGF16s Red1 - N1 after transplanted into rats with traumatic brain injury. Methods To transfect neural stem cells with Lipofectamine 2000. PBS, neural stem cells and gene- transfected neural stem cells were respectively transplanted into rats following traumatic brain injury, the expression of gene- transfected cells were checked by RT- PCR in vivo,neurological function was checked in all groups after transplantation by NSS. Result Gene- transfected cells could express VEGF165 in vivo, the values of NSS in C group were significantly lower than A group from 3 d after transplantation, B group were significantly lower than A group from 7 d after transplantation. Conclusion The gene - transfected neural stem cells could reduce brain injury by secreting VEGF165.
