摘    要:目的探讨NR2B特异性拮抗剂Ro25-6981对大鼠脑缺血-再灌注(I-R)后海马齿状回颗粒下层(SGZ)神经发生的作用。方法 64只SD大鼠随机均分为对照组(A组)、假手术组(B组)、脑I-R组(C组)和Ro25-6981 0.6、1.2、2.4、4.8和9.6μg/kg组(分别为D1、D2、D3、D4和D5组)。术后7d取脑,各组大鼠处死前2h侧脑室注射5-溴脱氧尿嘧啶核苷(BrdU),免疫组织化学染色观察SGZ中BrdU阳性细胞变化。结果与A、B组相比,C组SGZ的BrdU阳性细胞数明显增加(P<0.01);而D1、D2、D3、D4和D5组BrdU阳性细胞数均较C组减少(P<0.05或P<0.01)。结论Ro25-6981能抑制大鼠脑I-R后海马齿状回SGZ神经发生。

Abstract:Objective To investigate the effect of NR2B specific antagonist Ro25-6981 on the neurogenesis in the subgranular zone(SGZ) of the hippocampal dentate gyrus of rats after cerebral ischemia-reperfusion(I-R). Methods Sixty-four SD rats were equally randomized into eight groups of A (normal control),B(sham operated),C(cerebral I-R model),D1(treated with Ro25-6981 0.6 μg/kg),D2(treated with Ro25-6981 1.2 μg/kg),D3(treated with Ro25-6981 2.4 μg/kg),D4(treated with Ro25-6981 4.8 μg/kg) and D5(treated with Ro25-6981 9.6 μg/kg).On the 7th day after operation,the brain was taken and BrdU was injected into lateral ventricle at 2 hours before sacrifice.The positive cells of BrdU in the SGZ were detected by immunohistochemistry. ResultsCompared with groups of A and B,the positive cells of BrdU in the SGZ were significantly increased in group C(P<0.01).While the positive cells of BrdU in groups of D1,D2,D3,D4 and D5 were all lower than those in group C(P<0.05 or P<0.01). Conclusion Ro25-6981 inhibits the neurogenesis in the SGZ of the hippocampal dentate gyrus of rats after cerebral I-R.
