Cytotoxicity of Benzo(a)pyrene-7,8-dihydrodiol on Genetically Enginered V79 Chinese Hamster Cells Expresing Rat Liver Cytochrome P450 1A1

摘    要:本文采用细胞计数和中性红摄取法观察了BP7,8diol对XEM2(稳定表达了大鼠肝细胞色素4501A1)和V79(未表达P4501A1)细胞生长的作用,目的为建立体外细胞毒模型。结果表明,在1.0、2.5、5.0μmol·L1不同浓度下,BP7,8diol对XEM2细胞产生很强的毒性,其毒性具有浓度和时间依赖性,在相同的浓度下BP7,8diol对V79细胞并没有影响,同时也发现,细胞色素P450酶抑制剂αNF对BP7,8diol引起的细胞毒具有保护作用。这些结果说明,BP7,8diol对XEM2细胞毒性是通过细胞色素P4501A1的作用引起的。
