凋亡调控通路P53-MDM2-P14ARF对结肠癌生物学行为的影响

摘    要:采用免疫组织化学 S- P法检测 3 6例正常结肠组织、 2 8例结肠腺瘤及 42例结肠癌组织中凋亡调控蛋白P53、 MDM2 、 P1 4 ARF三者的表达。P53和 MDM2 在结肠癌中的表达明显高于正常结肠组织 ( P <0 .0 5 ) ,P1 4 ARF在结肠腺瘤和结肠癌中的表达明显低于正常结肠组织 ( P<0 .0 5 )。P53和 MDM2 在结肠癌中的表达与分化程度及临床分期无明显关系 ( P >0 .0 5 ) ,而 P1 4 ARF在低分化及 C期结肠癌中的表达明显低于高分化及 A期结肠癌 (P <0 .0 5 )。三者的相关分析提示 ,P53与 MDM2 之间为正相关 ( r =0 .63 3 9) ,与 P1 4 ARF之间为负相关 ( r =-0 .9169) ,MDM2 与 P1 4 ARF之间亦存在负相关 ,但相关不显著 ( r=- 0 .3 2 68)。认为 P53、 MDM2 和 P1 4 ARF在人结肠组织中的表达异常可能与结肠癌的多种生物学行为有关 ,三者联检的协同表达可作为结肠癌辅助诊断和判断预后的参考指标

Abstract:The apotosis regulatory proteins expression of P 53,MDM 2 and P 14ARF were determined by immunohistochmical staining in clinical specimens of 36 cases of human normal colon and 28 cases of colonic adenomas and 42 cases of colonic adenocarcinomas mucosal lesion.The positive staining rates of P 53 and MDM 2 in adenocareinomas were higher than that in normal mucosa (P<0.05),The positive staining rates of P 14ARF in adenomas and adenocarcinomas were lower than that in normal mucosa (P<0.05).The expressions of P 53 and MDM 2 in variant differentiation and clinic stage of colorectal cancer were not different (P>0.05),but the expression of P 14ARF in low differentiation and C stage of adenocarcinomas was lawer obvious than that in high differentiation and A stage of colorectal cance(P<0.05).Their correlation analysis suggusted that there were positive correlation between P 53 and MDM 2 (r=0.6339),negative correlation between P 53 and P 14ARF(r=-0.9169).There was no correlation between MDM 2 and P 14ARF(r=-0.3268).These suggest that P 53,MDM 2 and P 14ARF are expressed in human colorectal cancer,which might contribute to various biological behaviors of the cancer.United detection of P 53,MDM 2 and P 14ARF might be acted as an assistant marker of colorectal cancer.
