摘    要:用两次胰蛋白酶消化,共10次离心洗涤的方法,自感染弓形体小鼠的腹水内提纯弓形体速殖子。经台盼蓝染色、间接荧光抗体法检测与电镜观察,检测虫体活力、抗原性及形态变化。试验前,弓形体的台盼蓝自然着色(死亡)率是6%。末次离心洗涤后,胰蛋白酶消化组的弓形体有65%被台盼蓝染色;水洗对照组为41%。两组有显著性差异(P< 0.01)。用弓形体阳性人血清(滴度1:16),检测两组弓形体,与正常对照虫体相比,其荧光着色特点与滴度均未见明显改变。透射电镜发现,胰蛋白酶消化组与水洗对照组的弓形体,大多数的质膜呈波浪形并有一些疣状突,尤其是胰蛋白酶消化的虫体,其类锥体端的质膜肿胀、增厚,层次消失。

Effect of trypsinase digestion on tachyzoites of Toxoplasma gondii
Abstract:The peritoneal fluid of mica infected with Toxoplasma gondii was treated twice with double trypsinase digestion and normal Saline centrifugal washing for ten times to purify tachyzoites.Before treatment,6% of the tachyzpites-of Toxoplasma gondii were naturally stained(dead ).But following the final washing,65% of the parasites were stained in group of trypsinase digestion,41% of that in the control group The difference is highly significant (P<0.01).However,no change was found in feature and titre of IFA of the two groups.During the testing,human serum of anti-Toxoplasma gondii with titre 1:16 was used with normal parasite as control.Electron microscopic studies show that the plasmolemma of most tachyzoites treated by trypsinase or normal saline become wavy and some warty projections appear.
