肺耐药相关蛋白表达与NP方案治疗非小细胞肺癌疗效的关系

摘    要:目的:研究非小细胞肺癌(NSCLC)患者肿瘤组织中肺耐药相关蛋白(lung resistance-related protein,LRP)表达与患者化疗疗效的关系。方法:利用实时荧光定量RT-PCT及Western blot法,对42例初治Ⅲ、Ⅳ期NSCLC患者化疗前肿瘤组织活检标本中的LRPmRNA基因扩增及检测LRP表达,并评价NP方案(长春瑞滨十顺铂)化疗对NSCLC患者的疗效,探讨LRP基因表达与NP方案治疗非小细胞肺癌疗效关系。结果:LRP基因表达在肿瘤组织中显著增强(P〈0.05);18例部分缓解(PR)和稳定(SD)患者肿瘤组织中LRP表达明显低于进展(PD)患者的表达(P〈0.05);在PR与SD患者肿瘤组织中,LRP表达也存在着差异,但两者之间差异无显著性(P〉0.05)。结论:NSCLC患者肿瘤组织中LRP表达与患者对化疗疗效有关,提示LRP的表达可作为预测NSCLC化疗敏感性的指标。

Abstract:Objective: To investigate the relationship between expression of lung resistance- related protein(LRP) gene in non-small lung cancer (NSCLC) biopsies and its sensitivity to vinorelbine (NVB) combined with cisplatin(DDP). Methods:The real time quantitative RT-PCR and Western blot methods were administered to detect the amplification and expression level of mRNA of LRP gene, 42 NCSCL patients accepted chemotherapy of NP regimen and the responses were recorded. Results: Forty-two patients were treated with 2 to 4 cycles NP regimen. There were significant difference of the expression level of mRNA of LRP gene between carcinoma tissues and lung tissues(P〈0.05); 18 patients showed responses to the chemotherapy with partial response(PR) rate and stead disease(SD), the expression level of mRNA of LRP gent were significantly lower than pa tients with progressive disease(PD) ; the expression Ievel of rnRNA of LRP gene between patients with PR was lower than of patients with SD, but the difference was no significant(P〉0.05). Conclusion: The expression level of the LRP gene in NSCLC had relationship with the sensitivity of cytotoxic drugs. Patients with high level of LRP gene expression would have poor re- sponse to chemotherapy.
