摘    要:目的应用蛋白质组学技术分析肝癌血清中差异表达蛋白的临床意义。方法应用表面加强激光解析电离飞行时间质谱(SELDI-TOF-MS)技术,对肝癌、肝硬化、慢性乙型肝炎及健康对照组血清蛋白质进行检测,应用Ciphergen公司软件对蛋白质波谱进行分析。结果质荷比(m/z)为11492.92的蛋白质波峰强度仅肝癌组明显增强,与患者的肝功能、AFP等均无相关性。以波峰的信号噪声比(S/N)大于5为标准,该蛋白质诊断肝癌的灵敏度为50.8%,特异性为95.6%。结论质荷比(m/z)为11492.92的蛋白质可能是肝癌的生物学标志物,对AFP阴性患者的实验室诊断可能具有辅助作用。

Abstract:Objective To analyze differential expression protein by proteomic technology in hepatocellular carcinoma(HCC).Methods Serum protein from patients with HCC,liver cirrhosis (LC),hepatitis and healthy controls were detected by surface-enhanced laser desorption-ionization time-of-flight mass spectrometry(SELDI-TOF MS).Ciphergen software was used to identify proteomic features.Results The peak intensity of 11 492.92 Da protein was significantly higher in patients with HCC than in others.No significant correlation was found between the peak intensity and AFP value or hepatic function.The method had a sensitivity of 50.8% and a specificity of 95.6% with the criterion of S/N>5.Conclusion The peak of 11 492.92 Da protein is a potential biomarker for the diagnosis of HCC.It is likely a complementary method for the diagnosis of HCC with negative AFP.
