P物质和血管活性肠肽在慢传输型便秘大鼠肠壁内的变化和可复性研究

摘    要:目的观察便秘模型大鼠小肠和结肠P物质(SP)和血管活性肠肽(VIP)的变化及自然恢复情况,了解大黄所致便秘的可恢复性。方法Wistar大鼠60只,按照完全随机的方法分为便秘组、恢复组、对照组各20只,便秘组和恢复组给予大黄建立便秘大鼠模型,恢复组于建立模型完成后任其自然恢复6周。以放射免疫方法和免疫组织化学方法测定比较各组大鼠小肠和结肠VIP和SP的变化。结果与对照组比较,便秘组和恢复组大鼠的小肠、结肠SP含量均显著降低(P<0.05),恢复组小肠SP含量显著高于便秘组(P<0.05),恢复组结肠SP含量与便秘组比较无差别(P>0.05)。便秘组小肠和结肠VIP表达较对照组明显降低(P<0.05);恢复组VIP表达较便秘组高(P<0.05),但低于对照组(P<0.05)。结论长期应用大黄等蒽醌类泻剂对肠道神经系统有明显的损害作用,小肠和结肠SP含量降低,VIP反应性降低,停用大黄恢复性饲养6周后肠道神经递质VIP和SP部分恢复。

Abstract:Objective To observe the changes of substance P(SP)and vasoactive intestinal peptide(VIP)in the small intestine and colon of Wistar rats with constipation and to investigate the possibility of natural recovery of constipation caused by Rhubarb in rats.Methods 60 healthy Wistar rats were randomly divided into control group,constipation group and recovery group,20 rats in each group.The rats in constipation group and recovery group were given Rhubarb to set up constipation models of rats.The rats in recovery group were recovered naturally for six weeks,the levels of VIP and SP in small intestine and colon were detected by immunohistochemical stain and radioimmunoassay.Results As compared with those of control group,the levels of SP in small intestine and colon in constipation group and recovery group were significantly decreased(P<0.05);the levels of SP in small intestine in recovery group were significantly higher than those of constipation group(P<0.05);there was no significant difference in the levels of SP in colon between constipation group and recovery group(P>0.05).The levels of VIP in small intestine and colon in constipation group were significantly higher than those in control group(P<0.05).The levels of VIP in small intestine and colon in recovery group were significantly higher than those of constipation group(P<0.05),however,which were obviously lower than those of control group(P<0.05).Conclusion The long-term administration of Rhubarb can damage enteric nervous system through changing the expression of VIP and SP in small intestine and colon,with decrease of content of SP and the reactivity of VIP,however,six weeks after stopping the administration of Rhubarb,the expression of VIP and SP may recover partly.
