摘    要:目的 探讨二血管阻断加低血压法建立大鼠全脑缺血再灌注模型制作方法的改良,以增加铸模的成功率.方法 21只SD大鼠随机分为实验组16只,对照组5只.实验组采用改良二血管加低血压法制模,对照组不做任何处理;对实验结果进行统计,计算出存活例数和死亡例数之后开颅取大脑组织进行形态学检查,再根据检查结果统计出造模成功率.结果 16只大鼠中均无死亡现象,2只缺血效果不佳,造模成功率87.5%.经HE染色后可观察到实验组与对照组相比脑组织出现大量神经元坏死,胞浆浓缩,核固缩现象.结论 改良的二血管阻断加低血压法可以有效的降低造模的死亡率,能够较好的建立大鼠全脑缺血再灌注模型,对脑复苏的进一步研究提供良好实验基础.

Improvement of two vessel occlusion plus hypoteusion method in global brain ischemia model of rats

Abstract:Objective To establish the global cerebral ischemia/reperfusion model by the means of two vessel occlusion add hypotension, study changes of the global cerebral structure in rats. To provide conditions for the research of further nerve protection in the recovery of brain. Methods Establish the global cerebral is- chemia-reperfusion model in 16 SD rats by two-vessel occlusion plus hypotension. Calculate the number of alive cases and death cases, and make a craniotomy for cerebral tissue from morphological observation at the end of the experiment. Results 14 rats modeling success of the 16 rats, whose success rate was 87.5%. Obvious patho- logical lesions of nerve cells including neuronal necrosis, concentrated neuronal cytoplasma and karyopycnosis are observed in global cerebral ischemia-reperfusion rats. Conclusion The two vessel occlusion plus hypoten- sion method has a steady effect and a lower mortality of modeling. It can better establish global cerebral ischemi- a/reperfusion model of in rats, which can provide a good experimental basis for the recovery of the brain re- search.
