大孔树脂分离纯化半枝莲总黄酮的研究

摘    要:目的研究不同型号大孔树脂纯化半枝莲总黄酮的工艺条件及参数。方法以静态饱和吸附量、洗脱量、静态洗脱率为考察指标,比较了5种大孔树脂,并对AB-8树脂的动态比上柱量、比吸附量、比洗脱量进行了考察,以总黄酮转移率和纯度为指标对树脂吸附工艺条件进行了筛选。结果所比较的5种树脂中,AB-8型树脂具有最佳的吸附及洗脱参数,其最佳工艺是用浓度为3.12mg·mL^-1,pH=2.0的上样液,以3倍柱体积去离子水,12倍柱体积70%乙醇依次洗脱,总黄酮转移率为80%左右,其纯度为60%左右。结论AB-8树脂综合性能较好,适合于半枝莲中总黄酮的纯化。

Abstract:Objective To investigate the technology for adsorbing and separating total flavanoids from Scutellaria barbata with different types of macroporous resin (MR). Methods The static capacity adsorption, static elution ratio of 5 types of MR were studied respectively and compared to find the optimum one. The dynamic saturation ratio and dynamic absorption ratio of AB-8 and the yield, and purification degree of total flavanoids were researched and compared to obtain the optimum technological parameters. Results The AB-8 type of MR possessed optimum adsorption and elution parameters. The concentration of Scutellaria barbata sample extract was 3.12 mg · mL^-1 and its pH was 2.0. After being eluted with 3 multiple column volume of distilled yield of total flavanoids was 80% and the product purity was comprehensive adsorption, and it could be used to purify the water, 12 multiple column volume of 70% ethanol, the 60%. Conclusion The AB-8 type of MR showed better extract of Scutellaria barbata.
