活化蛋白激酶在骨肉瘤中的表达及其意义

摘    要:目的]探讨细胞分裂素活化蛋白激酶(MAPK)中细胞外信号调节蛋白激酶(ERK)、c-jun氨基末端激酶(JNK)、P38三条通路在骨肉瘤发生中的作用。方法]用免疫组织化学技术EnVision^TM法检测48例骨肉瘤标本及25例成骨性良性肿瘤标本中ERK、JNK、P38蛋白的表达,并比较他们的差异。结果]ERK、JNK、P38三种蛋白在骨肉瘤组的阳性率分别为83.3%(40/48),72.9%(35/48)和85.4%(41/48),在成骨性良性肿瘤组中的阳性率分别为16.0%(4/25),12.0%(3/25)和20.0%(5/25),骨肉瘤组织中ERK、JNK、P38蛋白阳性率及阳性强度均明显高于成骨性良性肿瘤(P〈0.01)。结论]MAPK中ERK、JNK、P38三条通路在骨肉瘤发生中均发挥了重要作用,用免疫组织化学方法检测MAPK,可为临床骨肉瘤与良性骨肿瘤的诊断与鉴别诊断提供参考指标。

Abstract:Objective]Extracellular signal-activated kinases(ERK),c-Jun N-terminal kinase(JNK) and P38 signal transduction pathway were three members of the MAPK family which mediated cellular proliferation,differentiation and apoptosis.The purpose of this study was to investigate the effects of ERK,JNK and P38 MAPK on pathogenesis and progression of human osteosarcoma.Method]EnVisionTM immunohistochemical technique was used to detect the expressions of ERK,JNK and P38 protein among the 48 osteosarcoma and 25 osteoblastic benign tumor samples,and their differences were compared.Result]The positive rates of ERK,JNK and P38 protein were 83.3%(40/48),72.9%(35/48) and 85.4%(41/48) respectively in 48 osteosarcoma,and 16.0%(4/25),12.0%(3/25) and 20.0%(5/25) in 14 osteoblastic benign tumor,the statistical analysis of the positive rates were performed using analysis of variance with P<0.01 shown statistical difference.Their positive rates and expressive intensities in osteosarcoma were higher than those in osteoblastic benign tumor.Furthermore,The results showed the localization of ERK,JNK and P38 protein to both the nucleus and to cytoplasm,ingeneral,those located in nucleus were more than that in cytoplasm.Conclusion]These findings indicated that the three signaling transduction pathway of MAPK played an important role in osteosarcogenesis.Detecting the three protein of tumor cells by immunohistochemistry may provide helpful references for differential diagnosis of osteosarcoma and osteoblastic benign tumor.
