摘 要: | 目的 研究 2型糖尿病 ( T2 DM)患者脂蛋白脂酶 ( L PL)基因位点变异对大血管病变易感性的影响。方法 选择 2 16例 T2 DM患者 ( T2 DM组 ) ,采用 PCR-RFL P检测其第 6、第 8内含子基因多态性 ,并与 63例正常对照组比较。结果 ( 1)对照组与 T2 DM组 L PL/Pvu 、Hind 基因多态性频率分布相似 ( P>0 .0 5 ) ;( 2 ) P-P-型大血管病变发生率为 62 % ,明显高于 P+P+型 ( 3 6% ) ( P<0 .0 5 ) ,而且 P-P-型与大血管病变呈正相关 ( r=+1.67,P=0 .0 0 8) ;( 3 ) H+ H+型大血管病变发生率为 45 % ,高于 H-H-型 ( 2 2 % ) ,但无统计学意义 ( P>0 .0 5 ) ;( 4 ) H+H+型三酰甘油 ( TG)水平明显高于 H-H-型 ( P<0 .0 5 ) ,P+P+型 TG和低密度脂蛋白胆固醇 ( LDL -C)水平分别明显低于和高于 P-P-型 ( P<0 .0 5 )。结论 LPL /Pvu 基因多态性调控 T2 DM患者并发大血管病变的危险性

Relationship between DNA polymorphism at the lipoprotein lipase gene and macroangiopathy in patients with type 2 diabetes mellitus |

Abstract: | Objective To study the effect of variation at the lipoprotein lipase (LPL) gene locus on the susceptibility of macroangiopathy in patients with type 2 diabetes mellitus (type 2 DM). Methods PCR RFLP technique was used to determine the DNA polymorphism of the 6th and 8th intron of lipoprotein lipase gene in 216 patients with type 2 DM (type 2 DM group). The results were compared with 63 healthy subjects (control group). Results (1) The prevalence distribution of lipoprotein lipase gene polymorphism of PvuⅡ/HindⅢ are similar between control group and type 2 DM group( P >0.05). (2) The prevalence of macroangiopathy in P P genotype group is 62%, which are significantly higher than 36% in P+P+ genotype group ( P <0.05). Moreover P P genotype is positively correlation with macroangiopathy ( r = +1.67, P =0.008). (3) The prevalence of macroangiopathy in H+H+ group (45%) is slightly higher than that in H H group(22%), but there was not statistical differences between H+H+ group and H H group ( P >0.05). (4) The triglyceride (TG) level in H+H+ group is significantly higher than that in H H group ( P <0.05). Meanwhile the TG and low density lipoprotein cholesterol (LDL C) levels in P+P+ group are significantly lower and higher than those in P P group, respectively ( P <0.05). Conclusion The risk of macroangiopathy in type 2 DM patients may be regulated by LPL/PvuⅡgenotypes. |
