| 摘 要： | 采用自制抗辣根过氧化酶单克隆抗体(抗HRP McAb)进行了鼠单克隆PAP(PAPm)研制。并对其染色特性进行观察。以抗HRP McAb AP_4制备PAPm、酶与抗体克分子比值以1:3为佳;腹水与上清制备的PAPm免疫组化染色效果相似,ELISA显色前者低于后者;用于肝组织HBsAg定位检测,本室AP_4、AP_2、AP_5三种抗HRP腹水制备PAPm的染色效价,显色强度及本底显色相当,均接近或达到Sigma公司同类制品的效果。为采用McAb进行临床免疫组化研究提供了可靠的试剂保证。

| Abstract: | PAP complexes have been devloped by using the mouse monoclonal antiperoxidase antibody and HRP. The different conditions of preparing PAP complexes were studied, and the optimal conditions was established. The mouse monoclonal PAP complexes have been used in immunohistochemical study and ELISA test. The experimental results show that the specific staining titration, background staining intensity of self-prepared PAP complexes are satisfactory. |
