慢性髓性白血病和急性淋巴细胞白血病细胞中Wnt信号通路活性的差异

摘    要:目的:探讨慢性髓性白血病(CML)和急性淋巴细胞白血病(ALL)细胞中Wnt信号通路活性是否存在差异.方法:利用慢病毒基因转染法在经BCR-ABL融合基因诱导形成的CML和ALL细胞中建立Wnt信号通路报告系统BAT-RED,检测2种白血病细胞中Wnt信号通路活性;PCR方法检测2种细胞中BAT-RED基因整合情况;通过向培养细胞中添加氯化锂,人为增强Wnt信号活性,反向验证Wnt信号通路活性在2种细胞中存在差异.结果:PCR结果显示2种白血病细胞都稳定整合了Wnt信号通路报告基因BAT-RED,在42%(188/448)的慢性粒细胞白血病细胞中可以检测到Wnt信号通路被激活,而ALL细胞仅散在少数细胞中可以检测到Wnt信号通路报告基因BAT-RED被激活;添加氯化锂后,ALL细胞中Wnt信号通路活性增强.结论:CML细胞中Wnt通路活性明显高于ALL细胞,提示Wnt信号通路在2种类型白血病细胞的增殖和演进过程中具有不同作用.
