黄芪、党参提取物抑制肺癌细胞诱导血管内皮细胞迁移的实验研究

摘    要: 目的 观察黄芪、党参提取物对肺癌细胞诱导的血管内皮细胞迁移的影响,探讨扶正中药抗肿瘤生长的机制.方法 采用免疫组化和图像分析技术,观察不同浓度的黄芪、党参提取物对人小细胞肺癌细胞(NCI-H446)VEGF蛋白表达的影响.采用Matrigel invasion chamber共培养法,观察黄芪、党参提取物对NCI-H446诱导人血管内皮细胞(ECV - 304) 迁移的抑制作用.结果 黄芪、党参提取物浓度达到40%(V/V)能够抑制NCI-H446诱导的ECV-304细胞迁移,并且具有显著的剂量-效应关系(r=-0.73,P=0.01).浓度达到50%(V/V)时可以明显抑制NCI-H446细胞VEGF表达(P=0.02).结论 高浓度的黄芪、党参提取物能够抑制肺癌细胞诱导的血管内皮细胞迁移,这种抑制作用可能与其抑制肺癌细胞VEGF表达有关.

Abstract:Objective To investigate the effect of Astragalus and Cedonopsis extract (ACE) on the migration of endothelial cells induced by lung cancer cells in vitro .Methods Cells of human umbilical vein endothelial cell line ( ECV- 304) and human small cell lung cancer cell line (NCI - H446) were cocultured in Matrigel invasion chamber system, the effect of ACE on the migration of ECV- 304 induced by NCI- H446 was investigated. The VEGF expression in NCI- H446 was examined by immunohistochemical staining and image analysis.Results ACE at concentration of over 40% (V/V) significantly inhibited the chemotactic migration of ECV - 304 cells induced by NCI - H446 cells in a dose - dependent manner( r = -0.73, P =0.01).The expression of VEGF in NCI - H446 cells was reduced after treatment with ACE in concentration of 50% (V/V)( P =0. 02).Conclusions ACE inhibits the endothelial cell migration induced by lung cancer cells and VEGF expression in lung cancer cells , suggesting that high dosage ACE can suppress angiogenesis and has possible therapeutic action on cancer.
