参附汤萃取液成分对阿霉素致心衰大鼠血流动力学、心肌自噬及凋亡的影响

摘    要:目的观察参附汤萃取液成分对阿霉素致心衰大鼠血流动力学、心肌自噬及凋亡的影响。方法建立阿霉素致大鼠心力衰竭动物模型,采用16导生理记录仪同步记录左室内压(LVSP),左室内压最大上升速率(+dp/dtmax),左室内压最大下降速率(-dp/dtmin),采用SYBR Green I法测定自噬相关基因Beclin-1,Atg5mRNA表达量及凋亡相关基因Caspase mRNA的表达量。结果连续给药6 W后,空白组与模型组比较左心室内压、左心室内压最大上升速率及左心室内压最大下降速率有明显降低,说明模型成立;参附汤水煎液组、参附汤萃取液成分组、参附注射液组与模型组比较,左心室内压、左心室内压最大上升速率及左心室内压最大下降速率有显著升高(P0.01);与空白组比较,模型组自噬相关基因Beclin1、Atg5 mRNA、凋亡相关基因Caspase mRNA的表达量均明显上升(P0.01);与模型组比较,参附汤水煎液组、参附汤萃取液成分组、参附注射液组自噬相关基因Beclin1、Atg5 mRNA、凋亡相关基因Caspase mRNA的表达量均明显下降。结论参附汤萃取液成分能改善阿霉素致心衰大鼠血流动力学相关指标,抑制心肌细胞自噬和凋亡。

Abstract:Objective to observe the effect of Shen Fu Decoction extraction liquid components on adriamycin induced heart failure rats hemodynamics, myocardial autophagy and apoptosis. Methods to establish the animal model of heart failure induced by adriamycin in rats, using 16 polygraph record left ventricular pressure ( LVSP), maximum rising rate of left ventricular pressure ( + dp/dtmax), left ventricular pressure ( - dp/dtmin), the largest decline rate by SYBR Use Green I method to Determine autophagy related gene Beclin - 1, the amount of AtgSmRNA expression and apoptosis related gene Caspase mRNA. Results continuous medicine administration for 6W, the blank group compared with the model group, the internal pressure of left ventricle, left ventricular pressure and maximum rate rose and the left ventricular pres- sure maximum falling rate had decreased, the model was established; Shen Fu Decoction group, Shen Fu Decoction extract group, Shen Fu injection group compared with the model group, the internal pressure of left ventricle, left ventricular pressure and maximum rate rose and left ventricular pressure maximum falling rate was significantly increased (P 〈 0.01 ) ; compared with the blank group, model group, the expression of autophagy related gene Beclinl, Atg5, mRNA, apoptosis related gene Caspase and mRNA were significantly increased ( P 〈 0.01 ) ; compared with the model group, Shen Fu Decoction group, the expression of Shen Fu Decoction extract group, Shen Fu injection group of autophagy related gene Beclinl, AtgS, mRNA, apoptosis related gene Caspase and mRNA decreased obviously. Conclusion Shen Fu Decoction extraction liquid components can improve the hemodynamics in rats with heart failure induced by adriamycin related index, inhibition of autophagy and apoptosis of myocardial.
