内皮素-1对前列腺癌PC3细胞系的侵袭、迁移能力影响

目的探讨内皮素1对前列腺癌PC3细胞系的侵袭、迁移能力的影响,并初步研究其机制。方法体外培养PC3细胞并以内皮素1干预,通过划痕实验、Boydenchamber细胞侵袭和迁移实验测定PC3细胞侵袭、迁移能力;RTPCR和明胶酶谱法分别测定PC3细胞基质金属蛋白酶(MMP)2、MMP9的mRNA表达和蛋白合成、激活的水平。结果与对照组比较,内皮素1组PC3细胞的侵袭、迁移能力显著提高,合成和激活MMP2、MMP9水平显著提高(P<0.05)。结论内皮素1能提高前列腺癌细胞的侵袭、迁移能力,促进MMP2、MMP9合成和激活是其机制之一。

Effects of Endothelin-1 on the Migration and Invasion Abilities of Prostate Carcinoma Cell Line PC3

Purpose To explore the effects of Endothelin-1 on the migration and invasion abilities of prostate carcinoma cell line PC3. Methods PC3 cells were cultured in vitro and interfered with endothelin-1.The migration and invasion abilities of PC3 cell were observed by erasion trace test,Boyden-chamber chemotaxis and chemoinvasion assay.The mRNA expression of MMP-2 and MMP-9 were detected by RT-PCR synthesis,and activation of MMP-2 and MMP-9 were detected by gelatin zymography,respectively. Results Compared with control group,the migration and invasion abilities of PC3 cell of eodothelin-1 group were statistically higher,the synthesis and activation of MMP-2 and MMP-9 of eodothelin-1 group were statistically higher,P< 0.05. Conclusions Endothelin-1 promotes the migration and invasion abilities of prostate carcinoma cells,and one of the mechanisms is to promote the synthesis and activation of MMP-2 and MMP-9.
