膜联蛋白 A1 蛋白在异丙酚对脓毒症大鼠肠保护作用中的机制

摘    要:目的观察膜联蛋白 A1(Annexin A1)蛋白在异丙酚对脓毒症大鼠肠保护作用中的机制。方法采用大鼠股静脉注射脂多糖复制脓毒症模型,按完全随机化原则分为 6 组:正常对照组、内毒素组、异丙酚组、脂肪乳组、内毒素+异丙酚组、内毒素+脂肪乳组。酶联免疫吸附法(ELISA)检测肠组织中肿瘤坏死因子(TNF-α)、白细胞介素-1(IL-1β)、白细胞介素-1(IL-6)的表达; Western Blot 检测肠组织中 Annexin A1 表达;HE 染色比较各组肠组织形态。结果与对照组相比,脂多糖组 IL-1β,IL-6,TNF-α 表达明显升高,Annexin A1 表达明显降低;脂多糖+异丙酚组 IL-1β,IL-6,TNF-α表达明显降低,Annexin A1 表达明显升高(P< 0.05)。结论异丙酚对脓毒症大鼠肠道有明显的保护和抗炎作用,且该作用可能与膜联蛋白 A1 蛋白相关。

Abstract:ObjectiveTo investigate the protection of Annexin A1 on the injury of intestinal protective effect of propofol in rats with sepsis. MethodsSepsis of rat model was induced by injection of lipopolysaccharide in rat femoral vein. The rats were randomized to 5 groups: normal control group(Group C), LPS group(Group L), propofol group(Group P), LPS+Propofol group (Group L + P), Intralipid group(Group I), LPS + Intralipid group(Group L + I). The intestinal tissue in each group were stained with hematoxylin and eosin(HE) and examined under microscope. The expression of tumor necrosis factor(TNF-α), interleukin1(IL-1) and leukocyte mediated endothelin-1(IL-6) on intestinal tissue were detected by ELISA. Western blot was used to detect the expression of Annexin A1. ResultsCompared with the control group, the expression of IL-1, IL-6, TNF-α was significantly increased, the Annexin A1 was obviously decreased in group L, while the opposite in group L+P. ConclusionPropofol may have protective and anti-inflammatory effects on the intestinal tract of rats with sepsis, and the effect may be correlated with the Annexin A1.
