高效液相色谱波长切换法测定老慢清糖浆中异嗪皮啶、迷迭香酸和柚皮苷的含量

目的:建立同时测定老慢清糖浆中异嗪皮啶、迷迭香酸和柚皮苷含量的液相色谱测定法。方法:色谱柱为Waters SunFire^TM C₁₈ (4.6 mm×150 mm, 5 μm);以乙腈-0.1%磷酸水溶液(18:82)为流动相,流速为1.0 mL·min^-1,柱温:30℃,检测波长:0~12 min为342 nm;12~18 min为284 nm; 18~35 min为342 nm。结果:嗪皮啶进样量在0.019~0.335 μg范围内线性关系良好(r=0.9998, n=6),平均回收率为99.57%,RSD为1.5%(n=9);迷迭香酸进样量在0.022~0.397 μg范围内线性关系良好(r=0.9999,n=6),平均回收率为99.55%,RSD为1.3%(n=9)。柚皮苷进样量在0.085~1.531 μg范围内线性关系良好(r=0.9999,n=6),平均回收率为99.19%,RSD为1.5%(n=9)。结论:该方法准确、快捷、可靠,可更好地用于老慢清糖浆中异嗪皮啶、迷迭香酸和柚皮苷的质量控制。

Simultaneous determination of contents of isofraxidin,rosmarinic acid and naringin in Laomanqing syrup by using HPLC wavelength switching method

OBJECTIVE To establish an HPLC wavelength switching method to simultaneously determine contents for isofraxidin, rosmarinic acid and naringin in Laomanqing syrups. METHODS Waters SunFire^TM C₁₈ (4.6 mm×150 mm, 5 μm) column was used. Mobile phase was acetonitrile-water containing 0.1% phosphoric acid (18:82) at a flow rate of 1.0 mL· min^-1. Detection wavelength was 342 nm (0-12 min) for isofraxidin, 284 nm (12-18 min) for naringin and 342 nm (18-35 min) for rosmarinic acid. RESULTS Isofraxidin had a good linearity in the range of 0.019-0.335 μg, the average recovery was 99.57% with RSD of 1.5% (n=9). Rosmarinic acid had a good linearity in the range of 0.022-0.397 μg, the average recovery was 99.55% with RSD of 1.3% (n=9). Naringin had a good linearity in the range of 0.085-1.531 μg, and the average recovery was 99.19% with RSD of 1.5% (n=9). CONCLUSION The method is simple, accurate and reproducible. It can be used to determine contents of isofraxidin, rosmarinic acid and naringin simultaneously in Laomanqing syrups under the same chromatogram conditions.
