宁夏枸杞总黄酮对恶性胶质瘤细胞株U87-MG表型的影响

摘    要:目的:研究宁夏枸杞总黄酮对人恶性胶质瘤U87-MG细胞增殖、迁移以及侵袭的影响。方法:采用贴壁法培养人恶性胶质瘤细胞。实验分为空白对照组(不加任何处理)和黄酮处理组(20,40,80,160 mg·L-1)。用CCK-8法测定不同浓度宁夏枸杞总黄酮作用U87-MG细胞不同时间后,细胞的增殖变化情况;RT-PCR和real-time PCR法检测肿瘤细胞内增殖细胞核抗原(PCNA)mRNA水平的表达;免疫细胞化学(immunocytochemistry,ICC)检测PCNA蛋白表达;Transwell和细胞划痕实验检测细胞的迁移能力;Transwell法检测细胞侵袭能力。结果:与对照组相比,处理组的U87-MG细胞增殖被抑制,且呈一定的时间-剂量依赖关系;处理组的PCNA mRNA和蛋白水平含量均降低,迁移以及侵袭能力减弱;且上述指标的变化随枸杞总黄酮浓度的增加而趋于明显。结论:宁夏枸杞总黄酮能够抑制恶性胶质瘤细胞的增殖、迁移以及侵袭。

Abstract:OBJECTIVE To study inhibitory effects of total flavones of Lycium barbarum L.on proliferation,motility and invasion of human glioma cell lines.METHODS U87-MG cells were cultured by tissue sticking method.The experiment had assigned blank control group (no-treatment) and flavones group (20,40,80,160 mg·L-1).IC50 was assayed by CCK-8.Proliferation of U87-MG cells were observed with CCK-8 at different concentrations and times.mRNA levels of intracellular proliferating cell nuclear antigen (PCNA) was measured by RT-PCR and real-time PCR.Expression of PCNA was performed by immunocytochemistry (ICC).Tumor cell motility was studied by a wound-healing model and transwell in vitro.Migration of tumor cells were investigated by transwell method.RESULTS Compared with control group,proliferation in flavones groups was obviously reduced in a time-dose dependent manner.Expression of mRNA and protein of PCNA,cell migration and invasion of treatment groups were obviously reduced by increasing total flavones.CONCLUSION Total flavones of Lycium barbarum L.can suppress proliferation,migration and invasion of human malignant glioma cells.
