盐知母降血糖活性部位作用研究

摘    要:目的: 对前期筛选出的盐知母活性部位的降糖作用进行深入研究,进一步证明其降血糖作用,为后期研发提供依据。方法: 采用HPLC法,以PNPG(对硝基苯基α-D-吡喃葡萄糖苷)为底物测定α-葡萄糖苷酶的反应产物PNP(对-硝基酚)的浓度,比较盐知母不同降血糖活性部位(乙酸乙酯层萃取物、三氯甲烷层萃取物)对α-葡萄糖苷酶的抑制作用;采用脂肪乳加腹腔注射STZ(链脲佐菌素)诱导2型糖尿病大鼠模型,并随机分组,灌胃14 d后测定空腹血糖值(FBG),并用试剂盒测定胰岛素、血清三酰甘油、总胆固醇等水平。比较盐知母不同降血糖活性部位对2型糖尿病大鼠降血糖效果。结果: 盐知母降血糖活性部位均对α-葡萄糖苷酶有抑制作用,且盐知母三氯甲烷层萃取物优于乙酸乙酯层萃取物;与模型组相比,各给药组均显著改善FBG、OGTT、HbAlc、Homa-IR等水平及TC、TG等血脂水平。结论: 盐知母三氯甲烷层萃取物对α-葡萄糖苷酶有明显抑制作用,对2型糖尿病大鼠具有降血糖和降血脂作用,是盐知母降血糖的活性部位。

Abstract:OBJECTIVE To investigate effects of hypoglycemic effects of active fractions pre-screened in salt processed Anemarrhena Rhizoma (SPAR), and provide scientific basis for later research and development. METHODS PNPG (p-nitrophenyl α-D-glucopyranoside) was used as substrate to determine concentration of p-nitrophenol (PNP), reaction product of α-glucosidase, by using HPLC, and inhibiting effects of different active fractions of SPAR on α-glucosidase were compared. Type 2 diabetes rats were induced by intraperitoneally injecting STZ and fat, and randomly divided into several groups. After gastric gavage for 14 days, fasting blood glucose (FBG) was determined, and test kits were used to determine levels of insulin, serum triglycerides and total cholesterols. Hypoglycemic effects of active fractions of SPAR were compared.RESULTS Different active fractions of SPAR all had inhibiting effects against α-glucosidase, and chloroform extract was superior to ethyl acetate extract. Compared to control group, FBG, OGTT, HbAlc, Homa-IR, triglycerides and total cholesterols were all improved significantly.CONCLUSION Extract of SPAR by chloroform is active fraction with hypoglycemic effects, can inhibit α-glucosidase activity obviously, and improve effects against hypoglycemia and dyslipidemia in rats with type 2 diabetes.
